目的 观察马来酸罗格列酮对雌性大鼠骨代谢的影响。方法 3月龄雌性Wistar大鼠40只随机分为4组,正常对照组(NC组)、低剂量(3mg.kg-1.d-1)罗格列酮组(Ros-1组)、中剂量(10mg.kg-1.d-1)罗格列酮组(Ros-2组)和高剂量(30mg.kg-1.d-1)罗格列酮组(Ros-3组)。每日定时按照每公斤体重6ml灌胃,NC组给予0.5%羧甲基纤维素钠溶液灌胃作为对照,干预12周。处死后,测定右侧股骨骨密度、血清骨代谢标记物和相关激素及细胞因子水平。结果 Ros-3组的股骨骨密度较其他三组显著降低,反映骨形成的血清骨钙素水平在Ros-2及Ros-3组显著降低,而其他骨代谢标记物无显著变化。各组的雌二醇、睾酮、瘦素水平无差异。Ros-3组IGF-1水平降低。各组炎症因子的水平无有意义的变化。结论 较大剂量的罗格列酮能减少正常雌性大鼠的骨形成,降低骨密度,对循环中雌二醇、睾酮及多数细胞因子无显著性影响。 |

Abstract:0bjective To investigate the effects of rosiglitazone on the bone metabolism in female rats.Methods Forty 3-month-old female Wistar rats were randomly distributed into 4 groups,and were treated with rosiglitazone maleate at doses of 3,10,and 30mg kg-1.d-1(Ros-1,Ros-2,and Ros-3),or 0.5% carboxymethycellulose(NC)by gavage(6ml kg-1.d-1)for 12 weeks. Bone mineral density(BMD)of the fight femur,selum bone metabolic markers,and related hormones and eytokines were measured after the rats were killed.Results BMD of the femur in group Ros-3 decreasad significantly compared to that in 3 other groups.The serum levels of osteoealein decreased significantly in groups Ros-2 and Ros-3,but no change occurred in other bone metabolic markers.There was no difference in the 1evels of estradiol.testosterone,and leptin among the groups.The serum level of IGF-1 decreased in group Ros-3.No significant change occurred in the inflammatory factors among the groups.Conclusion Rosiglitazone at a higher dose decreases bone formation and BMD in the normal female rats.Rosiglitazone has no influence in serum levels of estradiol,testosterone,and most cytokines. |
