      目的 探讨大鼠骨关节炎模型与正常对照组软骨退变基因表达差异,从基因网络角度探讨骨关节炎的发病机制。方法 通过切断大鼠右膝交叉韧带、内侧副韧带,建立骨关节炎动物模型,采用代表28000个不同基因的Genomic Instrumentation Services公司芯片检测骨关节炎和正常大鼠右膝关节软骨的基因表达,对差异基因表达进行生物信息学分析。结果 大鼠骨关节炎软骨退变有显著差异表达的基因406个。其中305个基因上调,101个基因下调。差异表达基因涉及生长因子、发育、肥胖、能量代谢、信号传导、免疫等方面的基因群。结论 骨关节炎软骨退变基因表达谱存在软骨细胞凋亡相关的基因表达异常,信号传导过程异常、免疫相关基因的表达下调、多个转录因子的普遍上调等特征,为研究骨关节炎在基因水平上的病理变化提供了基础。
英文摘要:
      Objective To investigate the difference of degenerative cartilage gene expression profile Between arthritic rats and normal controls.And to explore the pathogenesis of osteoarthritis from gene network view.Methods Arthritic animal model was established by cutting off cruciate ligament and medial collateral ligament of the right knee joint.The gene expression in arthritic or normal articular cartilage of rat right knee was measured using a gene chip by Genomic Instrumentation Services.The differential gene expression was analyzed based on information biology.Results A total of 406 differentially expressed genes were identified in degenerative rat arthritic cartilage,and among them there were 305 up-regulated genes and 101 down-regulated genes.Differentially expressed genes involved gene group of growth factor,growth,obesity,energy metabolism,signal transduction,and immunity.Conclusion Gene expression profile of degenerative osteoarthritic cartilage is characterized by differentially expressed genes related to chondrocyte apoptosis,signal transduction abnormality.Down-regulated immunity-related genes,and many up-regulated transcription factors.This provides a basis for researching osteoarthritis pathological change at gene level.
