| 目的建立NHA2基因敲出小鼠模型，观察NHA2一／一小鼠骨髓细胞分化为破骨细胞的能力
及生长表型变化。方法首先用NHA2+／一杂合子小鼠繁殖得到NHA2一／一纯合子小鼠；以小鼠尾
巴为材料做普通PCR鉴定其基冈型；用RT—PCR检测基囡敲出小鼠NHA2转录情况；从小鼠中取出骨
髓进行破骨细胞诱导，观察其分化能力。结果NHA2一／一小鼠骨髓细胞体外诱导成成熟破骨细胞
能力减弱；但小鼠生长表型无明显变化。结论NHA2作为盐离子载体在体外对破骨细胞的成熟分
化有一定影响，在小鼠体内可能与其它离子载体共同调节盐平衡系统。 |
| 英文摘要: |
| 0bjective To establish NHA2一／-mice model。and to observe the differentiation ability of bone
marrow cells to osteoelasts and the growth phenotype．Methods NHA2+／一mice were bred to NHA2一／·
mice．Mouse tails were used to test the gene phenotype using regular PCR method．RT—PCR wss used to
detect the NHA2 transcription by NHA2·／-mice．Mice bone wits separated to do X-ray scanning and to lest
BMD．Bone marrow celh were extracted from the mice to induce osteoclasts and to observe their
differentiation ability．Results NHA2·／-mice model WaS established successfully．11le result of RT·PCR
indicated that there was He NHA2 transcription in NliA2-／一mice．The X-ray showed that BMD of NHA2一／一
mice writs slightly higher．and the skull bigger than that of the wild·type mice．The ability of osteoclast
differentiation was decreased in NHA2·／-mice in vitro．But the phenotype旷NHA·／-mice WaS not changed，
Conelusion NHA2 as an iron carrier Call affect the differentiation and maturation of osteoelasts iu vitro．It
may interact with other iron carriers to regulate the iron balance． |
