Objective To observe the effect of tanshinone II A on the expression of metalloprotease and its inhibitor in osteoblasts from human femoral head and to explore its function for the treatment of femoral head necrosis. Methods Human femoral head osteoblasts were cultured in vitro. IL-1β was added to facilitate the expression of MMP and TIMP in osteoblasts. The expression of MMP and TIMP were measured with qPCR in the control group,including 3 samples,and measured 3 times repeatedly. Tanshinone IIA was add?ed in the experimental group to perform the same measurement. The results were compared. Results There were no augmentation of MMP8,MMP12,MMP15,and TIMP4 in qualitative PCR.The expressions of MMP1,MMP2,MMP3,MMP7,MMP9,MMP13,MMP14,MMP17,TIMP1,and TIMP3 in the experimental group were significantly different comparing with those in the control group using qPCR ( P <0. 05 ). The expressions of MMP10,MMP11,and TIMP2 in the expermiental group were not significantly different comparing with those in the control group ( P > 0. 05 ). Conclusion There is no expression of MMP8,MMP12,MMP15,and TIMP4 in osteoblasts from human femoral head. Tanshinone IIA can effectively inhibit partial secretion of MMP in osteoblasts from human femoral head and accelerate partial secretion of TIMP to postpone femoral head necrosis. |
