目的观察骨质疏松症大鼠骨骼、骨骼肌I型胶原蛋白表达变化的影响,探讨中医防治骨质疏 松症的作用机制。方法将120只雌雄各半的大鼠随机分为正常对照组、模型对照组、补肾中药组、 健脾中药组、活血化瘀中药组和骨疏康中药组6个组。采用Western blot方法测定大鼠骨骼、骨骼肌 I型胶原蛋白表达。结果①各组大鼠骨组织均可检测到I型胶原的蛋白表达。与正常组比较,各 组大鼠骨组织I型胶原的蛋白表达均明显下降(!<0.01);与模空组比较,各治疗组大鼠骨组织I型 胶原的蛋白表达均明显升高,其中补肾组、骨疏康组升高趋势最显著(P<0. 01 )。②各组大鼠骨骼肌 均可检测到I型胶原的蛋白表达。与正常组比较,各组大鼠骨骼肌I型胶原的蛋白表达均明显下降 (P<0.01);与模空组比较,各治疗组大鼠骨骼肌I型胶原的蛋白表达均明显升高(!<0.01),其中补 肾组升高趋势最明显。结论①骨质疏松症的形成,可能与骨骼、骨骼肌I型胶原表达的异常变化有 关。②补肾、健脾方法通过调控骨质疏松症大鼠的骨骼、骨骼肌I型胶原蛋白表达,对骨质疏松症具 有一定的防治作用。 |

Objective To explore the mechanism of traditional Chinese herb in treatingosteoporosis by investigating protein expression of collagen I ( COL I) in skeleton and skeletal muscle of osteoporotic rats. Methods One hundred and twenty rats ( male/female = 1 : 1 ) were randomly divided into 6 groups as following : blank control group, model control group, invigorating the kidney group, invigorating the spleen group, promoting blood flow group, and Gusukang group. The protein expression of COL I in skeleton and skeletal muscle was tested using Western-blotting. Results 1 ) Protein expression of COL I could be detected in the skeleton of all the groups. Compared to blank control group, the expression in all other groups decreased significantly ( P <0.05 ) . Compared to model control group, the expression in all the herbal treating groups elevated significantly,among which the expression in invigorating the kidney group and Gusukang group elevated most significantly ( P < 0. 01 ) . 2 ) Protein expression of COL I could be detected in skeleton muscle of all the groups. Compared to blank control group, results in all other groups decreased significantly ( P < 0. 05 ) . Compared to model control group,expression in all herbal treating groups elevated significantly, among which the expression in invigorating the kidney group elevated most significantly ( P < 0.01). Conclusion 1) The development of osteoporosis may be associated with abnormal expression of COL I in the skeleton and skeletal muscle. 2 ) Treatments of invigorating the kidney and invigorating the spleen show its efficacy to prevent osteoporosis by regulating the protein expression of COL I in the skeleton and skeletal muscle. |
