      目的 通过观察三组不同发育阶段雌性豚鼠后肢膝关节软骨降解的形态学和生理特征,探讨雌性豚鼠自发性骨性关节炎软骨形态学变化与体重和蛋白多糖及雌激素的关系。方法 选用Hartley 雌性豚鼠15只,分为出生后30天、90天、210天共3个年龄组,每组各5只,取胫骨膝关节软骨。经HE染色在光学显微镜下,观察3组豚鼠膝关节软骨降解的形态学变化,同时测定每组豚鼠的体重及血清生化指标蛋白聚糖和雌二醇水平。结果 全部5只30天组雌性豚鼠胫骨关节软骨组织结构层次分明,关节面光滑完整,没有降解现象;全部5只90天组雌性豚鼠胫骨关节软骨都发生了轻度以上的降解损害;全部5只210天组雌性豚鼠胫骨关节软骨都发生了 重度降解的形态学改变。蛋白聚糖 随年龄增加明显下降,雌二醇的浓度随着年龄的增长而持续升高。体重在90天组明显增加。以上数据各组间差异均有非常显著性意义(P<0.01 )。结论 雌性豚鼠自发性骨关节炎的发生与血清蛋白聚糖的下降或体重的增加有关,而可能与血清雌激素水平无关。

      Objective To observe the morphology and physiological features of hind limb knee cartilage degeneration in female guinea pigs at different development stages in 3 groups, and to evaluate the relationship among cartilage morphological changes, body weight,and proteoglycans and estradiol in female guinea pigs with spontaneous osteoarthritis ( OA ). Methods Fifteen female Hartley guinea pigs were selected. They were divided into 3 groups according to different age: 30,90, and 210 days after born ( n =5 in each group). The medial tibial plateau cartilage was collected. After HE staining, optical microscope was used to observe the morphological changes of knee joint cartilage degeneration of guinea pigs in 3 groups. Body weight and serum biomarkers such as proteoglycans and estradiol in each group were measured. Results The tibial articular cartilage of all 5 female guinea pigs at the age of 30 days had distinct structure and smooth surface,and no sign of degeneration was observed. At the age of 90 days,mild or more than mild degeneration of the articular cartilage was observed. At the age of 210 days,morphological changes due to severe degeneration of the articular cartilage were observed. The serum proteoglycan level decreased significantly with the age increased (P < 0. 01 ). The serum concentration of estradiol increased continuously along with the increase of age. Body weight in the group at the age of 90 days increased significantly (P <0. 01 ). Conclusion The pathogenesis of spontaneous osteoarthritis in female guinea pigs may relate to the decrease of serum proteoglycan level or the increase of body weight, but may have no relation with serum estradiol level.
