目的测定绝经后妇女血清基质金属蛋白酶40 (MMP40)和金属蛋白酶抑制物4(TIMP4)水平,并探讨其与骨密度 数值和骨代谢指标的关系。方法选取武汉地区120名48岁至65岁绝经后女性,用酶联免疫吸附试验(ELISA)测定血清 MMP40、TIMP4以及整合素(Integrin)?2p1、雌二醇(E2)、骨保护蛋白(OPG)、骨保护蛋白配体(OPGL)的水平,计算MMP- 10/TIMP4比值,用双能X线吸收法(DEXA)测定腰椎正位,股骨颈,华氏区和大粗隆的骨密度(丽D)。同时,按照WHO标准 将人选女性分为骨密度正常组、低骨量组和骨质疏松组。结果①骨质疏松组中血清MMP40检测结果(644. 25 ± 19. 21)pg/ ml 高于正常组(347. 08 ±21. 41)pg/ml (P <0.05),而 TIMP4 检测结果(134 ± 116XL 低于正常组(146 ± 130)哗//。②骨 质疏松组中血清MMP40与骨密度、血清E2、OPGL和Integrin ?2^1测定结果存在明显负相关性(P <0. 05),和OPG和CTX 存在明显正相关性(P < 0. 05)。结论血清MMP40和MMP40/TIMP4比值与绝经后骨质疏松症妇女骨密度和骨代谢指标 具有关联性。两者可能通过整合素途径参与绝经后骨质疏松症骨代谢高转换过程。 |

Objective To determine the serum levels of matrix metalloproteinase-L0 (MMP-L0) and tissue inhibitor of matrix metalloproteinase-L (TIMP--) in postmenopausal women,and to explore the correlation of the serum levels of MMP--3 and TIMP- 1 with the bone mineral density (BMD) and bone metabolic markers. Methods One hundred and twenty postmenopausal women in Wuhan area,aging from 48 to 65 years old,were selected. The serum levels of MMP-10,TIMP--,integrin o2p1,E2,OPG, and OPGL of all the subjects were detected using ELISA. And the ratio of MMP-1 ^^TIMP-- was calculated. BMD of the lumbar vertebrae,the femoral neck,the Ward ’ s area,and the greater trochanter was detected using dual energy X-ray absorptiometry (DEXA). Then,all the selected subjects were divided into 3 groups according to the WHO criteria: the normal control group,the low bone mass group,and the osteoporosis group. Results The serum concentration of MMP-10 in osteoporosis group (644. 25 士 19. 21 pg/ml) was significantly higher than that in normal control group (347. 08 士 21. 41 pg/ml,P <0. 05),while the serum concentration of TIMP-- was lower in osteoporosis group (134 ±116 jxg^L) than that in normal control group (146 士 130 pg/L). In osteoporosis group,the notable negative correlations between MMP-10,and BMD,serum E2,OPGL,and integrin o2p1 were observed (P < 0. 05). Positive relations were observed between MMP-40,OPG,and CTX (P < 0. 05). Conclusion The serum level of MMP-10 and the ratio of MMP-40/TIMP-- are significantly correlated with BMD and bone metabolic markers in postmenopausal osteoporosis women.
