      目的研究绝经后骨质疏松症肾阳虚证的关联基因LTBP1及PCSK6的mRNA表达水平,探讨绝经后骨质疏松症肾阳 虚证与LTBP1及PCSK6基因表达的关联性。方法随机选择绝经后骨质疏松症患者,中医辨证肾阳虚证组25例,32例健康 绝经后妇女为对照组,RT孑CR法检测外周血LTBP1及PCSK6 mRNA的表达。结果肾阳虚证组LTBP1 mRNA表达水平低 于健康对照组(F = 7.473,P = 0.008),差异有统计学意义;肾阳虚证组PCSK6 mRNA表达水平较健康对照组比较,有降低趋 势,但差异无统计学意义;通过GO分析参与到的生物途径有细胞外组织形成、代谢过程、磷酸化作用、蛋白磷酸化、TGFH3的 细胞外基质合成、BMP信号通路、细胞分泌等;通过Pathway分析参与到弹性纤维形成、细胞外基质组织、整合胰腺癌的通路、 TGFH3信号通路、发育生物学、神经生长因子信号转导等通路相关。结论骨质疏松症肾阳虚证与LTBP1基因表达下调相关 联。

      Objective To investigate the mRNA expression of LTBP1 and PCSK6 in postmenopausal osteoporosis patients with the kidney yang deficiency,and to explore the correlation among them. Methods The postmenopausal osteoporosis patients were randomly selected. After syndrome differentiation,25 patients were divided into the kidney yang deficiency group. And 32 healthy postmenopausal women were selected in healthy control group. The expression of LTBP1 and PCSK6 mRNA in peripheral blood was detected using real-time RT-PCR. Results The expression level of LTBP1 mRNA in the kidney yang deficiency group was lower than that in healthy control group (F = 7. 473,P = 0. 008). Compared with that in healthy control group,the expression of PCSK6 mRNA in the kidney yang deficiency group showed a declining trend,but no significant difference was observed. The results of GO analysis showed that the related biological pathways involved extracellular matrix organization, metabolic process, phosphorylation process,protein phosphorylation,synthesis of TGF beta in extracellular matrix,BMP signaling pathway,and cell secretion. According to the Pathway analysis,the postmenopausal osteoporosis with the kidney yang deficiency was mainly related to the formation of elastic fiber, extracellular matrix organization, the integrated pancreatic cancer pathway, TGF beta signaling pathway,developmental biology,and NGF signaling pathway. Conclusion The postmenopausal osteoporosis with the kidney yang deficiency is correlated with the down-regulated expression of LTBP1 gene.
