目的 通过观察六味地黄丸对绝经后骨质疏松症肾阴虚证JAK/STAT信号通路基因的影响,探讨六味地黄丸治疗绝经后骨质疏松症肾阴虚证的分子机制。方法 随机选择绝经后骨质疏松症肾阴虚证组6例,健康绝经后妇女对照组3例。六味地黄丸治疗肾阴虚证组3个月后,用JAK/STAT信号通路芯片检测其基因表达的变化。结果 治疗前肾阴虚证组与正常对照组相比,差异表达基因:PRLR、JUN、JUNB、INSR,表达均下调;肾阴虚证组治疗后与治疗前相比,差异表达基因有25条,上调 11条,下调14条,其中JUN、JUNB疗后明显上调。差异表达基因生物学功能分析,免疫相关基因:PRL、PRLR、OSM、ISG15、IL4R;细胞生长增殖与细胞周期相关基因:F2、SOCS3;与STAT蛋白相互作用的转录因子和共同活化基因:JUN、JUNB、 SMAD3、SMAD5、SMAW、SP1、YY1; JAK/STAT信号通路的负反馈调节基因:PIAS1、SOCS4;衔接蛋白基因:SRC、STAM。结论 六味地黄丸治疗绝经后骨质疏松症肾阴虚证的机制可能与其调控JAK/ STAT信号通路的相关基因有关,这些基因的功能与免疫调节、细胞生长增殖及成骨生长关联。 |

Objective To investigate the effect of Liuweidihuang pill on JAK/STAT signaling pathway gene expression in postmenopausal osteoporosis with the kidney yin deficiency,and to explore the molecular mechanism of Liuweidihuang pill in the treatment of postmenopausal osteoporosis with the kidney yin deficiency. Methods According to the TCM syndrome,6 women with the kidney yin deficiency and postmenopausal osteoporosis were randomly selected. Another 3 healthy postmenopausal women were selected as control group. Patients with the kidney yin deficiency were treated with Liuweidihuang pill for 3 months. The gene expression in JAK/STAT signaling pathway was detected using special chip. Results Compared with those in control group,the expression of 4 differential genes was down-regulated in the kidney yin deficiency group before the treatment,including PRLR,JUN,JUNB,and INSR. After the treatment,25 differentially expressed genes were identified in the kidney yin deficiency group. Among these 25 genes,11 genes were up-regulated,and 14 genes were down-regulated. The expression of JUN and JUNB was up- regulated significantly. According to the analysis of the biological function of the differentially expressed genes, the immune response genes included PRL,PRLR,OSM,ISG15,and IL4R. The cell growth and/or maintenance genes included F2 and SOCS3. The transcription factor or regulator genes that interact with STAT proteins included JUN,JUNB,SMAD3,SMAD5, SMAD4,SP1,and YY1. The negative regulators of the JAK/STAT pathway included PIAS1 and SOCS4. The SH3/SH2 adaptor genes included SRC and STAM. Conclusion The mechanism of Liuweidihuang pill in the treatment of postmenopausal osteoporosis with the kidney yin deficiency may be related to the regulation of the genes in JAK/STAT signaling pathway. These genes are involved in immunity function, cell proliferation differentiation, and the growth of osteogenesis. |
