      目的 通过构建乳鼠颅盖骨成骨细胞分离培养与鉴定方法,研究胰岛素样生长因子-l(Insulin-like growth factor-l,IGF- 1)及IGF4联合重组人甲状旁腺激素1-34( recombinant human parathyroid hormone 1-34,rhPTH1-34)对成骨细胞增殖及I型胶原蛋白mRNA表达的影响。方法 培养乳鼠成骨细胞,并观察其形态及功能,以原代培养乳鼠成骨细胞为实验模型,分空白组、PTH组、IGF4组及不同浓度IGF4作用的PTH介导的成骨细胞组(0、10、50、100 ng/L)。通过不同剂量的胰岛素样生长因子4(0、10、50、100 ng/L)联合10-9 mol/L重组人甲状旁腺素刺激体外培养的成骨细胞,用噻唑蓝(MTT)法检测细胞的增殖能力,RT-PCR法检测成骨细胞I型胶原蛋白mRNA的表达。结果 PTH和IGF4均可促进成骨细胞增殖;IGF4联合PTH可以促进成骨细胞增殖,且具有剂量依赖性。PTH联合IGF4使成骨细胞增殖能力增强、I型胶原蛋白mRNA表达增强。IGF4可以促进成骨细胞I型胶原蛋白mRNA的表达,且具有剂量依赖性。IGF4可以促进成骨细胞I型胶原蛋白mRNA的表达,且具有剂量依赖性。结论PTH与IGF4均可刺激成骨细胞增殖和分化,IGF4可促进PTH对成骨细胞的分化、增殖。两者合用其作用增强,有协同促进作用。

      Objective Using the rat calvaria bone osteoblast isolation and identification method,to study the effect of insulin-like growth factor-1 (IGF-1 ) and IGF-1 combined with recombinant human parathyroid hormone 1-34 (rhPTH1-34) on the osteoblast proliferation and the mRNA expression of type-1 collagen. Methods Osteoblasts from new born rats were cultured and their morphology and function were observed. Rat osteoblasts in primary culture were divided to blank control group,PTH group,IGF-1 group,and different concentrations of IGF-1 combined with PTH group (0,10,50,and 100 ng/L). The osteoblasts were stimulated using different concentrations of IGF-1 (0,10,50,and 100 ng/L) combined with 10-9 mol/L of rhPTH1 -34. Proliferation of the cells was assessed using MTT colorimetric assay and the mRNA expression of type-1 collagen was examined using RT-PCR. Results PTH and IGF-1 can both promote proliferation of osteoblasts. IGF-1 combining with PTH can promote osteoblast proliferation in a dose dependent manner. They together can increase the expression of type-1 collagen mRNA of the osteoblast. IGF-1 can promote osteoblast type 1 collagen mRNA expression in a dose dependent manner. Conclusion Both PTH and IGF-1 can stimulate the proliferation and differentiation of osteoblasts. IGF-1 can promote PTH-mediated osteoblast differentiation and proliferation. Using the two of them can improve the proliferation effect and they may have synergistic effect.
