Objective To evaluate the anti-osteoporosis effect of Eucommia ulmoides Oliv.and the effect of salt-roast processing product on the treatment efficacy in ovariectomized rats.Methods Forty female SD rats were divided into Sham group, OVX group, Eucommia ulmoides Oliv.group, and salt-roast processing Eucommia ulmoides Oliv.group.All the rats except in Sham group were ovariectomized.After 4 weeks of the operation, rats received oral Eucommia ulmoides Oliv.or its salt-roast processing product (4.0 g/kg?d) daily.The body weight of the rats was recorded.After a 12-week treatment, the serum calcium (S-Ca) and phosphorus ( S-P) were measured.Serum ALP, osteocalcin ( OCN) , and estrogen ( E2 ) were determined using ELISA kits.The uterus was weighed after execution of the rats and pathological examination was performed using HE staining.Bone mineral density of the bilateral femur was tested using dual-energy X-ray absorptiometry.Morphology of the trabecular and the number of the osteoblast were observed using HE staining.Microarchitecture of the trabecular bone was observed using micro-CT scan.Results Oral administration of Eucommia ulmoides Oliv.and the salt-roast processing product effectively inhibited ovariectomy-induced the body weight increase, increased S-Ca and E2 , decreased S-P, ALP, and OCN.They increased BMD of the femur, improved microarchitecture of the trabecular bone, and increased the number the osteoblast.The long-term use of the drug had no stimulating effect on the uterus.Conclusion Eucommia ulmoides Oliv.and the salt-roast processing product have good therapeutic effect on ovariectomy-induced osteoporosis in rats, and the effect of salt-roast processing product is better than unroasted drug. |
