      目的研究不同强度50Hz脉冲电磁场(Pulse electromagnetic fields, PEMFs)对小鼠成骨细胞系MC3T3-E1增殖与分化 的影响。方法 MC3T3-E1传代培养后分成七组,分别采用0. 0 mT、0. 6 mT、1. 2 mT、1. 8 mT、2. 4 mT、3. 0 mT和3.6 mT 50 Hz 脉冲电磁场处理,90 min/d。MTT法检测细胞增殖情况,成骨性分化检测处理3 d、6 d、9 d、12 d后ALP活性和首次处理12 h 后BMP-2、Runx-2、OPG基因表达情况。结果6种强度的脉冲电磁场均促进细胞增殖,其中0. 6 mT的促增殖效应最强(P < 0. 01)。在成骨性诱导培养条件下,细胞内ALP活性随电磁场处理时间的延长而逐渐增加,第9 d达到峰值,之后开始回落。 0. 6 mT、3. 0 mT和3. 6 mT均能提髙ALP活性,其中0. 6 mT始终处于最髙水平(P <0. 01)。三种基因的表达水平在首次处理 12 h,在0. 6 mT、1. 2 mT、3 mT和3. 6 mT组均显著提髙,但仍然是0. 6 mT的增强效应最为明显。结论0. 6 mT 50 Hz脉冲电 磁场具有最佳的促进MC3T3-E1细胞增殖和成骨性分化活性,这可能为采用脉冲电磁场治疗骨质疏松症提供了理想的治疗参数。

      Objective To investigate the effect of pulsed electromagnetic fields (PEMFs) on proliferation and differentiation of MC3T3-E1 cells. Methods MC3T3-E1 cells were divided into 7 groups after subculture. They were exposed to 50 Hz PEMFs (0.0,0. 6,1. 2,1. 8,2. 4,3. 0,and 3. 6 mT,respectively) for 90 min/day. Cell proliferation was assessed using MTT method. ALP activity was examined on 3,6,9,and 12 days. Expression levels of BMP-2,Runx-2,and OPG mRNA were analyzed after 12 hours of PEMFs. Results Six intensities of PEMFs promoted MC3T3-E1 cell proliferation. The highest promotion was achieved under 0. 6 mT intensity (P <0. 01). Under the osteogenic culture condition,ALP activity gradually increased along with the time of exposure PEMFs. The peak was on the 9th day and it decreased thereafter. ALP activity was promoted under 0. 6 mT,3. 0 mT,and 3. 6 mT of PEMFs exposure. The highest promotion was achieved under 0. 6 mT intensity (P <0. 01). The expression levels of the 3 genes increased after first 12 h PEMFs treatment at 0. 6 mT,1. 2 mT,3. 0 mT,and 3. 6mT intensity. The most significant effect was still at 0. 6mT exposure. Conclusion Exposure at 50Hz 0. 6mT of PEMFs significantly promotes the proliferation and differentiation of MC3T3-E1 cells. This may provide ideal reference for the treatment of osteoporosis using PEMFs.
