      目的 探讨长期使用糖皮质激素对大鼠的骨质量影响。方法 3月龄SD大鼠共38只,雌性。随机分成4组,分别是: (1)基础对照组(B组,8只,实验开始时处死,建立本批大鼠底线);(2)正常对照组(C组,10只,腹腔注射生理盐水75 mg/ kg);(3)低剂量激素组(L组,10只,腹腔注射甲强龙50 mg/kg) ;(4)高剂量激素组(H组,10只,腹腔注射甲强龙75 mg/kg)。 实验开始时,低剂量激素组和高剂量激素组连续使用甲强龙3天(75 mg/kg. d),停用一周后,按计划使用甲强龙,每周一次,用药24周后取股骨和第三腰椎行骨生物力学实验。结果(1)与正常对照组相比,低剂量激素组和高剂量激素组的实验后体重明显减少(P <0.05);(2)与正常对照组相比,低剂量激素组和高剂量激素组股骨的结构力学参数(载荷、刚性)和材料力学参数(破断强度、刚性模量)均显著降低(P <0. 05);(3)与正常对照组相比,低剂量激素组和高剂量激素组腰椎的材料力学参数(破断强度、刚性模量)均显著降低(P <0. 05)。结论 长期使用糖皮质激素激素,无论剂量高低,均会导致大鼠股骨和腰椎骨质量下降。

      Objective To determine the effect of long-term use of methylprednisolone on bone biomechanics of rats. Methods Thirty-eight 3 -month-old female SD rats were chosen. They were randomly divided into basal control group (B group,8 rats were sacrificed at the beginning of the experiment as baseline),normal control group ( C group, 10 rats were given intraperitoneal injection of 75mg/kg normal saline),low dose methylprednisolone group(L group,10 rats were given intraperitoneal injection of 50 mg/kg methylprednisolone),and high dose methylprednisolone groug (H group,10 rats were given intraperitoneal injection of 75 mg/kg methylprednisolone). Methylprednisolone was given in L group and H group for 3 days,and stopped for a week. Then the intraperitoneal injection was given once a week according to the plan. After 24 weeks of the treatment,the femurs and the lumbar spine L3 were collected for biomechanical experiment. Results (1) Compared to that in C group,the body weight in L group and H group decreased obviously after the experiment (P <0. 05). (2) Compared to those in C group,structural mechanical parameters ( load and rigidity) and material mechanical parameters ( breaking strength and modulus of rigidity) of the femurs in L group and H group significantly decreased (P<0. 05). (3) Compared to those in C group,structural mechanical parameters (load and rigidity) and material mechanical parameters ( breaking strength and modulus of rigidity) of the L3 in L group and H group significantly decreased (P <0.05). Conclusion The long-term use of methylprednisolone,regardless of dose,results in the decrease of bone quality of the femur and the lumbar spine of rats.
