目的探讨软骨细胞在肿瘤坏死因子(TNFa)作用下对其分泌基质蛋白及相关基因mRNA表达的影响。方法体外培 养的小鼠软骨细胞用25,50,100 ng/ml的hTNFa干预,不加人TNFa作为对照,通过RT-qPCR检测hTNFa对软骨细胞分泌基质 蛋白及相关基因mRNA表达的影响。结果hTNFa能降低软骨细胞分泌的n型胶原及刺激促n型胶原分泌的SOX9基因的表达; 对基质可提高基质酶MMP~9,MMP43,ADAMTS-4和ADAMTS-5基因的表达,提高炎性因子IL4a,TGFp,并增加凋亡基因BAX, caspase3的表达。结论hTNFa通过不同途径影响软骨细胞及基质相关基因的表达,从而促进了关节软骨的降解。 |

Objective To investigate the effect of tumor necrosis factor (TNFa) on the secretion of matrix protein and mRNA expression of related genes in cartilage cells. Methods Mouse chondrocytes were cultured in vitro and were additioned with 25, 50,and 100 ng/ml of hTNFa. No hTNFa cultures were as control. mRNA expression of related genes in chondrocytes was detected using RT-qPCR. Results hTNFa reduced type II collagen secretion in cartilage cells and stimulated SOX9 gene expression that stimulates pro-[I collagen secretion. It increased gene expression of MMP-9,MMP-L3, ADAMTS4, and ADAMTS-5 in the matrix. It also increased inflammation factors IL-La and TGFp,and increased apoptotic gene expression of BAX and caspase 3. Conclusion hTNFa affects the expression of matrix-elated genes in cartilage cells by different ways, thus stimulating the degradation of articular cartilage. |
