      目的 探讨绝经后骨质疏松症患者骨组织中类固醇受体辅助激活因子3(steroid receptor coactivator-3, SRC-3)和过氧化物酶体增殖物活化受体γ协同刺激因子-1α(peroxisome proliferator-activated receptor gamma coactivator 1α, PGC-1α)的表达及意义。方法 2012年6月~2013年9月,选择于广州军区广州总医院关节外科住院的行全髋关节置换患者20例,术前双能X线(DXA)骨密度仪测量患者腰椎1~4(L1-4)骨密度,根据WHO颁布的诊断标准,分为试验组(腰椎BMD T评分<–2.5,10例)和对照组(T评分>–1.0,10例),术中取一侧开路器切除的粗隆部位松质骨。荧光实时定量RT-PCR和Western blotting分别检测骨组织中SRC-3和PGC-1α等mRNA和蛋白的表达。结果 与对照组相比,试验组骨组织中SRC-3, PGC-1α, CBP/P300, P/CAF mRNA表达水平均下降(P值分别为<0.001, 0.036, 0.003, 0.027),OPN mRNA表达水平升高(P=0.004)。Osteocalin mRNA表达在试验组和对照组中无统计学差异(P>0.05)。试验组SRC-3和PGC-1α蛋白相对表达量较对照组明显下调,分别下调了(53.23±0.55)% (P=0.037)和(72.17±0.64)% (P=0.003)。结论 SRC-3和PGC-1α可能参与绝经后骨质疏松症发病的病理过程。

      Objective To explore the expression and significance of steroid receptor coactivator-3 (SRC-3) and peroxisome proliferator-activated receptor gamma coactivator 1α (PGC-1α) in the bone in patients with postmenopausal osteoporosis. Methods A total of 20 cases of postmenopausal women with osteoporosis, who were in the inpatient at orthopedic department in General Hospital of Guangzhou Military Command of PLA for total hip arthoplasty from June 2012 to September 2013, were selected. BMD of the lumbar spine (L1-4) was measured using DEXA before the operation. According to the WHO criteria. the subjects were divided to the experimental group (10 cases, T score < –2.5) and control group (10 cases, T score > –1.0). Cancellous bone samples were collected from the greater trochanter which was cut by open circuit device. The mRNA and protein relative expressions of SRC-3 and PGC-1α were determined using quantitative PCR and Western blotting, respectively. Results Compared with control group, the relative expression of SRC-3, PGC-1α, CBP/P300, and P/CAF mRNA in experimental group decreased (p<0.001, 0.036, 0.003, and 0.027, respectively), but the relative expression of OPN mRNA increased (p=0.004). No statistically significance was found in the relative expression of osteocalcin mRNA between the two groups. Compared with control group, the protein relative expression of SRC-3 and PGC-1αreduced significantly (53.23±0.55%, P=0.037; 72.17±0.64%, P=0.003, respectively). Conclusion SRC-3 and PGC-1α may be involved in the pathogenesis of postmenopausal osteoporosis.
