健骨方治疗膝骨关节炎疗效机制的实验研究

      目的 通过观察模型组、健骨方组及阳性组对大鼠骨关节炎软骨细胞中p38丝裂原活化蛋白激酶(p38 MAPK)表达水平的影响,确定膝骨关节炎的发病及修复机制与p38MAPK介导的细胞信号通路密切相关,通过在信号通路水平干预和调控 p38MAPK的表达水平,将有希望成为治疗膝骨关苷炎的新的有效途径。方法 选用SD大鼠40只,采用Hulth法制备大鼠膝骨关节炎模型,以健骨方灌胃为干预治疗手段,连续喂药3 月后处死大鼠,取出患侧关节软骨进行检测。Western blotting测定各组p38 MAPK表达水平。结果 Western blotting显示p38 MAPK磷酸化产物在模型组呈持续表达状态,且明显高于正常组,差异有统计学意义(P <0.05);在健骨方呈下降趋势表达状态,给药组与模型组比较,差异有统计学意义(P <0.05)。结论 通过实验研究p38MAPK信号传导通路与骨关节炎有密切关系,降低其表达水平能够对骨关节炎起到治疗作用;健骨方可影响p38MAPK信号传导通路,此研究阐明了健骨方治疗膝骨关节炎的疗效机制,为临床更加合理有效地治疗膝骨关节炎提供了理论依据。

      Objective To determine the mechanism of the association between the onset and repair of knee osteoarthritis and p38MAPK mediated cell signaling pathway, by observing the effect of the model group, Jiangu recipe group,and positive control group on expression of p38 MAPK in articular cartilage in osteoarthritis rats. It may hopefully become a new effective approach for the treatment of knee osteoarthritis through intervening and regulating the expression level of p38MAPK. Methods Knee osteoarthritis model was established in 40 SD rats using Hulth’s method. Jiangu recipe was fed intragastrically for intervention therapy and lasted for 3 months. The rats were killed the cartilage was extracted. The expression levei of p38MAPK was detected using Western blotting. Results Western blotting results showed that p38 MAPK in the model group continued to express and the expression was higher than that in the normal group significantly ( P < 0. 05). The expression of Jiangu recipe group decreased significantly (P < 0. 05) comparing to that in the model group. Conclusion p38MAPK signaling pathway has close relationship with osteoarthritis. The reduction of its expression level can treat osteoarlhritis. Jiangu recipe can affect p38MAPK signaling pathway.This study indicates the therapeutic mechanism of Jiangu recipe treating knee osteoarthrilis more reasonably and effectively.
