      目的 观察复方贞术调脂胶囊(FTZ)对糖皮质激素诱导骨质疏松大鼠股骨和腰椎骨密度及生物力学特性的影响。方法SPF级雄性SD大鼠32只,随机等分为4组:Nrm组为正常对照组,Met组为皮下注射甲强龙(Met) 5mg/(kg·d),每周5次,FTZL组和FTZH组在Met组基础上每日分别给予低剂量FTZ(1.5g/kg)和高剂量FTZ(6g/kg)灌胃,实验期为12w。QDR4500A型双能X线骨密度测定仪测定股骨和腰椎骨密度,MTS-858型生物力学实验机测定股骨和腰椎生物力学性能。结果 Met组大鼠股骨和腰椎的骨密度、最大载荷、刚度、弹性模量、皮质骨厚度、股骨中段截面面积和椎体横断面面积均显著低于Nrm组相应指标(Ρ<0.001, Ρ<0.01),股骨骨髓腔面积显著高于Nrm组(Ρ<0.01);FTZL组大鼠的股骨骨密度,最大载荷、刚度、弹性模量、皮质骨厚度、股骨中段截面面积和椎体横断面面积较Met组有升高趋势,股骨骨髓腔面积较Met组减少,但两组间各指标均无显著性差异(Ρ>0.05);FTZH组大鼠股骨和腰椎的骨密度、最大载荷、刚度、弹性模量、皮质骨厚度和椎体横断面面积显著高于Met组相应指标(Ρ<0.01, Ρ<0.05),股骨骨髓腔面积显著低于Met组(Ρ<0.05),股骨中段截面面积较Met组有增加趋势,但无显著性差异(Ρ﹥0.05),FTZH组腰椎的骨密度和最大荷载显著高于FTZL组(P﹤0.05),余各项指标间无显著性差异(Ρ﹥0.05)。结论 高剂量FTZ对改善糖皮质激素诱导骨质疏松大鼠股骨和腰椎的骨密度及生物力学特性效果显著。

      Objective To observe the effect of FTZ on BMD and biomechanics in the femur and lumbar vertebrae in glucocorticoid-treated rats. Methods Thirty-two male SD rats were randomly divided into 4 groups: age-matched normal control (Nrm), methylprednisolone group (Met, 5.0mg/kg, sc, per day for 5 days/week), FTZL group (gavage administration, 1.5g/kg/d), and Met plus FTZH gavage administration (6g/kg/d). The study period was 12weeks. BND of the left femoral diaphysis and the fifth lumbar vertebrae (L5) was evaluated using DXA. The biomechanics of the femur and L4 were tested. Results BMD of the left femur and L5, maximum load, rigidity, elastic modulus, cortical bone thickness, the middle sectional area in the femur, and the sectional area of the vertebrae were significantly lower in Met group than those in Nrm group (Ρ<0.001, Ρ<0.01). The marrow cavity area was significantly higher in Met than that in Nrm (Ρ<0.01). BMD of the femur, in maximum load, rigidity, elastic modulus, cortical bone thickness, the middle sectional area in the femur, and the sectional area of the vertebrae showed increasing trend in FTZL group comparing to Met group, and the marrow cavity area decreased, but with no statistical different (Ρ>0.05). BMD of the femur and L5, maximum load, rigidity, elastic modulus, cortical bone thickness, and the sectional area of the vertebrae were significantly higher in FTZH group than those in Met group (Ρ<0.001, Ρ<0.01). The marrow cavity area was significantly lower in FTZH than that in Met (Ρ<0.01). The middle sectional area in the femur showed increasing trend, with no statistical different (P>0.05). BMD of the L5 and maximum load in the L4 were significantly higher in FTZH than those in FTZL (Ρ<0.05). There was no significant difference in the rest of indicators between FTZH and FTZL (Ρ>0.05). Conclusion High dose of FTZ has active effect on BMD and biomechanics in the femur and lumbar spine in glucocorticoid-treated rats.
