青藤碱降低兔膝骨关节炎模型痩素含量及其受体表达

      目的 观察青藤碱关节腔注射对兔膝骨关节炎模型血清及关节液瘦素水平以及关节软骨瘦素受体(Ob-Rb)表达的影 响。方法 38只新西兰大白兔随机分为空白组、模型组、透明质酸钠组和青藤碱组。空白组不予任何处理,模型组、透明质酸 钠组和青藤碱组通过木瓜蛋白酶关节腔注射法诱导膝骨关节炎模型后,分别用生理盐水、透明质酸钠和青藤碱干预5周,双抗 体夹心酶联免疫吸附(ELISA)法检测血清及关节液瘦素含量,实时荧光定量PCR(qPCR)法检测Ob-Rb的mRNA表达, Western blot法检测Ob-Rb的蛋白表达。结果 模型组和透明质酸钠组血清、关节液瘦素水平较空白组均显著升髙(P < 0. 05),青藤碱组血清、关节液瘦素水平明显低于模型组(P <0. 01)和透明质酸钠组(P <0. 05);模型组Ob-Rb的mRNA和蛋 白表达较空白组均显著升髙(P <0.05,P <0.01),青藤碱组Ob-Rb的mRNA和蛋白表达明显低于模型组(P <0.05,P < 0. 01)。结论 青藤碱膝关节腔注射可降低兔膝骨关节炎模型血清及关节液瘦素水平,下调关节软骨Ob-Rb的表达。

      Objective To observe the effect of intra-articular injection of sinomenine (SIN) on leptin in serum and articular synovium as well as expression of leptin receptor (Ob-Rb) in rabbit knee osteoarthritis model. Methods A total of 38 New Zealand white rabbits were randomly divided into 4 groups: control group,model group,hyaluronate group,and sinomenine group. Knee osteoarthritis model was established with intra-articular injection of papain enzyme in model group,hyaluronate group,and sinomenine group and treatment with saline, sodium hyaluronate,and sinomenine,respectively,lasted for 5 weeks except for control group. ELISA method was used to detect the level of leptin in rabbit serum and articular synovium. qPCR was used to detect the mRNA level of Ob-Rb. Western blotting was used to detect the protein level of Ob-Rb. Results The level of leptin in serum as well as in articular synovium in model group was significantly higher than that in control group (P <0. 05),and the levels of serum and articular synovium leptin in sinomenine group was significantly lower than those in model group (P < 0. 01) and hyaluronate group (P <0. 05). mRNA and protein expression of Ob-Rb in model group were higher than those in control group (P <0.05,P <0. 01). mRNA and protein expression of Ob-Rb in sinomenine group were lower than those in model group (P <0.05, P <0. 01). Conclusion Intra-articular injection of sinomenine can reduce the level of leptin in serum and articular synovium and down-regulate the increased expression of Ob-Rb in cartilage in rabbit knee osteoarthritis model.
