补骨脂素对去势雌鼠E2、ERβ、TNF-α、IL-17的影响

      目的 观察补骨脂素对去势雌鼠E2、ERβ、TNF-α、IL-17的影响,探讨补骨脂素防治绝经后骨质疏松的机理。方法 选取3月龄雌性SD大鼠72只,随机分为假手术组、模型组、戊酸雌二醇对照组、壮骨止痛胶囊对照组、补骨脂素高剂量组、补骨脂素低剂量组,每组12只。除假手术组外,其它大鼠造成双侧去卵巢骨质疏松症病理模型,术后5d拆线后开始给药,连续13 周。最后一次给药后次日取血标本采用ELISA检测血清中雌激素(E2)、肿瘤坏死因子-α(TNF-α)、白细胞介素-17(IL-17〉的含量。提取左股骨组织mRNA样本进行RT-PCR反应检测骨组织TNF-α、ERβ、IL-17基因的表达。取右股骨脱钙切片免疫组化检测ERβ表达。结果 模型组去势雌鼠血清E2水平显著低于假手术组(P <0.01)。壮骨止痛胶囊显著升高血清E2水平 (P <0. 01),补骨脂素高剂量组血清E2水平显著高于模型组(P <0. 01),补骨脂素低剂量组血清E2水平与模型组相比无统计学差异(P >0.05)。模型组血清TNF-α、IL-17水平及股骨TNF-α、IL-17 mRNA表达显著高于假手术组(P<0.01),股骨ERβ显著低于假手术组(P <0.05)。戊酸雌二醇组血清TNF-α、IL-17水平及股骨TNF-α、IL-17 mRNA表达显著低于模型组,股骨ERβ显著高于模型组(P <0.05)。壮骨止痛胶囊组血清TNF-α、IL-17水平显著低于模型组(P<0.05),股骨TNF-α、IL-17mRNA表达非常显著低于模型组(P <0. 01),而股骨ERβ显著高于模型组(P <0. 05)。补骨脂素高剂量组血清TNF-α水平及股骨TNF- α mRNA表达显著低于模型组(P<0. 05),而血清IL-17水平及股骨IL-17mRNA表达非常显著低于模型组(P < 0. 01),股骨ERβ 显著高于模型组(P<0.05)。补骨脂素低剂量组血清TNF-α水平显著低于模型组(P <0.05),但其股骨TNF-α mRNA表达与模型组相比无统计学差异,而血清IL-17水平及股骨IL-17mRNA表达显著低于模型组(P <0. 05),股骨ERβ与模型组相比没有统计学差异。结论 补骨脂素可通过提髙去势雌鼠血清E2水平和股骨ERβ水平,降低去势雌鼠骨组织TNF-α、IL-17 基因表达和血清TNF-α、IL-17水平发挥抗绝经后骨质疏松症作用。

      Objective To investigate the anti-osteoporosis mechanism of psoralen by observing its effect on E2, ERβ, TNF-α, and IL-17 in ovariectomized rats. Methods Seventy-two 3-month-old SD female rats were randomly allocated into sham operation group, model group, estradiol group, Zhuanggu Zhitong ( ZGZT) capsule group, the high psoralen dosage group,and the low psoralen dosage group, with 12 rats in each group. Except the rats in the sham, operation group, the rats in the other groups were ovariectomized bilaterally. The medicines were given to rats via intragastic administration for 13 weeks. The levels of serum estradiol (E2), tumor necrosis factor alpha (TNF-α), and leukocyte mediated IL-17 (IL-17) were detected using ELISA method. The gene expressions of ERβ, TNF-α, and IL-17 in the left femurs were detected using RT-PCR. ERβ was detected in the right femurs with immunohistochemistry. Results The level of serum E2 in model .group was significantly lower than that in sham operation rats (P <0. 01). ZGZT capsule increased serum E2 significantly (P <0. 01). The level of serum E2 in the high psoralen dosage group was significantly higher than that in model group (P<0.01). It was no difference between the low psoralen dosage group and the model group (P> 0. 05). The levels of serum TNF-α and IL-17 and the gene expression of TNF-α and IL-17 in the femurs in the model group were significantly higher, but the expression of ERβ was lower than those in the sham operation group (P <0. 01). The levels of serum TNF-α and IL-17 and the gene expression of TNF-α and IL-17 in the femurs in the estradiol group were lower, but the expression of ERβ was higher than those in the model group (P <0. 01). The levels of serum TNF-α and IL-17 and the gene expression of TNF-α and IL-17 in the femurs in the ZGZT capsule group lower,but the expression of ERβ was higher than those in the model group (P < 0.01). The serum TNF-α and gene expression of TNF-α mRNA in the femur were lower in the high dosage of psoralen group than those in the model group, and the serum IL-17 and gene expression of IL-17 mRNA in the femur were even lower than those in the model group (P <0. 01). ERβ in the femur was higher than that in the model group. Serum TNF-α in low dosage of psoralen group was lower than that in the model group, but the expression of TNF-α mRNA was not different comparing with model group. Serum IL-17 and the expression of IL-17 mRNA in the femur were significantly lower than those in model group (P < 0. 05). The expression of ERβ in femur was not different comparing with the model group. Conclusion Psoralen can improve the levels of E2 and ERβ in ovariectomized rats. It also reduces the gene expression of TNF-α and IL-17 in the bone tissue and serum levels of TNF-α and IL-17,therefore exerts its anti-postmenopausal osteoporosis function.
