      目的 探讨壮骨方对糖尿病大鼠骨质疏松进程的干预作用及其对血清IGF-l、TNF-α水平的影响。方法 建立2型糖尿病大鼠模型,分为空白组、模型组、二甲双胍组及壮骨方高、中、低剂量6组,分别给予相应药物灌胃12周。12周后测定各组大鼠全身骨密度,行胫骨干骺端病理切片测定骨计量参数,采用酶联免疫吸附法测定各组血清IGF-1、TNF-α的水平。结果 模型组大鼠的骨密度、骨剂量参数、血清IGF-1水平均明显下降,血清TNF-α表达增高(P <0.01),壮骨方可提高血清IGF-1、 降低TNF-α水平,防止骨密度、骨参数下降(P <0. 01),且中药高剂量组作用均优于二甲双胍组(P <0. 01)。结论 壮骨方可能通过升高血清IGF-1水平、降低TNF-α水平,进而影响糖尿病骨代谢进程。

      Objective To study the effect of the traditional Chinese medicine,Zhuanggu-decoction,on serum IGF-1 and TNF-α in type 2 diabetes rats during the process of osteoporosis. Methods Rat type 2 diabetic model was established and the rats were divided into 6 groups:blank group, DM group, DM + metformin group (MET), DM + MET + low dose Zhuanggu-decoction (ZGF-L) group, DM + MET + medium dose Zhuanggu-decoction (ZGF-M) group, and DM + MET + high dose Zhuanggu- decoction (ZGF-H) group. The rats were gavaged with corresponding drugs for 12 weeks. After 12 weeks, the bone mineral density, quantitative bone histomorphometry of the tibial stem epiphyseal, and serum IGF-1 and TNF-α were measured in each group. Results The bone mineral density, quantitative bone histomorphometry parameters, and serum IGF-1 decreased, but serum TNF-α increased in DM group (P <0.01). Serum IGF-1 increased, serum TNF-α decreased, and the decrease of bone mineral density and histomorphometry parameters was prevented in Zhuanggu-decoction groups. The effect in DM + MET + ZGF-H group was better than that in DM + MET group (P < 0. 01). Conclusion Zhuanggu-decoction affects osteoporosis process by increasing serum IGF-1 and decreasing serum TNF-α level in type 2 diabetic rats.
