Objective To investigate the effect of anti-sclerostin antibody and treadmill exercise on bone remodeling in aged ovariectomized rats. Methods Female Wistar rats were randomly divided into 5 groups, Sham group, OVX group, OVX+E group, OVX+S group, and OVX+E+S group. After 14 weeks, samples were collected and the related indexes were tested. Results (1) Exercise alone reduced fat, increased the number of trabecular bone, reduced bone resorption marker NTx level, and prevented the loss of bone mineral density of the whole body and the femur in aged rats. (2) Scl-Ab alone increased bone formation marker OC, improved the cortical porosity, cortical bone volume, and cortical bone thickness, improved bone mineral density in ovariectomized rats. (3) The combination of exercise and the antibody reduced the amount of fat, improved bone mineral density and cortical bone parameters, increased bone mass and bone strength, improved OC level, and reduced NTx level. Conclusion Moderate treadmill exercise prevents bone loss in aged ovariectomized rats and improves bone health in ovariectomized rats. The treatment with anti-sclerostin antibody increases bone mass and bone strength, and prevents bone metabolism from the effect of ovariectomy in rats. The anti-sclerostin antibody and exercise show a good synergistic effect on bone remodeling in aged ovariectomized rats. |
