目的 研究补肾强督方对BMSCs膜内成骨中Wnt通路的作用。方法 BMSCs细胞分为空白对照组、西药对照组、补肾强督方低、中、高剂量组,进行膜内成骨诱导分化,ELISA法检测上清液ALP、BGP,Western blot和荧光定量PCR检测DKK-1和β-catenin蛋白和基因表达。结果 补肾强督方含药血清对BMSCs能促进BMSCs成骨分化中DKK-1蛋白和mRNA表达,抑制β-catenin蛋白和mRNA表达,且呈剂量依赖性,但对成骨能力无影响。结论 补肾强督方能调节BMSCs细胞成骨分化中Wnt通路,可能具有抑制AS骨化的作用。 |

Objective To observe the effect of Bushen Qiangdu recipe on Wnt/β-catenin pathway in intramembranous ossification by bone mesenchymal stem cells (BMSCs). Methods BMSCs were divided into the blank control group, the Western medicine group, the low-dose group of Bushen Qiangdu recipe, the middle-dose group of Bushen Qiangdu recipe, and the high-dose group of Bushen Qiangdu recipe. Intramembranous ossification was induced. Enzyme-linked immuno sorbent assay (ELISA) was used to detect supernatant alkaline phosphatase (ALP) and bone gla-protein (BGP). Protein and gene expression of DKK-1 and β-catenin was detected using Western blot and quantitative polymerase chain reaction (PCR). Results Bushen Qiangdu recipe-containing serum promoted the protein and mRNA expression of DKK-1 in osteogenic differentiation of BMSCs, and inhibited the protein and mRNA expression of β-catenin, in a dose-dependent manner. It had no effect on bone formation. Conclusion Bushen Qiangdu recipe regulates Wnt signaling pathway in osteogenic differentiation of BMSCs, and thus may inhibit the ossification in ankylosing spondylitis. |
