目的 探索甲状旁腺激素(PTH)对去势大鼠骨质疏松的防治作用。方法 30 只健康雌性 SD 大鼠随机行假手术(Sham,N =10)和双侧卵巢(OVX,N =20)切除手术后,所有 OVX 组大鼠随机的分成 2 组:OVX 组、PTH + OVX 组。术后第一天开始给予药物治疗,PTH + OVX 组:PTH 皮下注射(剂量 60 μg/kg,每周 3 次),直至手术后 12 周为止,实验截止时间之前所有大鼠注射碘海醇后处死取股骨行 Micro-CT 检测。结果 与 OVX 组比较,PTH + OVX 组股骨远端有较高的 BMD、BV/TV、
Tb. Th、Tb. N、Conn. D、血管体积分数和较低的 Tb. Sp,其中 Sham 组大鼠股骨远端有最高的 BMD、BV/TV、Tb. Th、Tb. N、Conn.
D、血管体积分数和最低 Tb. Sp。结论 甲状旁腺激素通过促进血管形成及骨量增加来防治去势大鼠骨质疏松。 |

Objective To investigate the effect of parathyroid hormone (1-34) on the prevention and treatment of osteoporosis inOVX rats. Methods After bilateral ovariectomy (OVX,n = 20) and sham operation (Sham,n = 10),all animals from OVX
group were randomly divided into two groups,OVX group and PTH + OVX group. Rats in PTH + OVX group received PTH (60μg/kg,3 times a week) for 12 weeks. The rats were injected iohexol to observe angiogenesis before sacrificed. The distal femurs
were evaluated using micro-computerized tomography. Results Comparing with OVX group,the femurs in Sham and PTH +OVX groups had a higher BMD,BV/TV,Tb. Th,Tb. N,Conn. D,and vascular volume fraction,but a lower Tb. Sp. Rats in
Sham group had the highest BMD,BV/TV,Tb. Th,Tb. N,Conn. D,and vascular volume fraction,and the lowest Tb. Sp.Conclusion Parathyroid hormone (1-34) prevents and treats osteoporosis through enhanced angiogenesis and osteogenesis. |
