      目的 研究左、右归丸及其拆方对去卵巢大鼠骨髓间充质干细胞(bone marrow stem cells,BMSCs)PPARγ、LPL、FABP4 mRNA的影响。方法 分别以空白对照组(CON)、模型组(OVX)、假手术组(SHAM)、左归丸组(ZGW)、右归丸组(YGW)、共同药组(GTY)、滋阴药组(ZYY)、补阳药组(BYY)和补佳乐组(BJL)共9组对BMSCs进行干预,MTT法检测各组BMSCs增殖情况;流式细胞仪鉴定BMSCs;用Q-PCR法检测过氧化物酶体增殖物激活受体γ(Peroxisome Proliferator-Activated Receptor γ, PPARγ),脂蛋白脂肪酶(lipoprotein,LPL),脂肪酸结合蛋白4(fatty acid binding protein 4,FABP4)mRNA的表达。结果 左、右归丸及其拆方均可显著下调BMSCs PPARγ、LPL、FABP4 mRNA的表达,其中YGW组可显著下调PPARγ、LPL 、FABP 4的mRNA(P<0.05)。结论 左、右归丸及其拆方均能抑制BMSCs的成脂分化,且“右归丸”抑制成脂分化的作用优于“左归丸”,以及纯阳药及二者的共同药组。

      Objective The present study was designed to evaluate the effects of Zuogui Wan and Yougui Wan and their disassembled prescriptions on PPARγ, LPL and FABP4 mRNA levels of bone marrow mesenchymal stem cells in ovariectomized rat. Method Serum of control Group (CON), ovariectomized group (OVX), sham group (SHAM), Zuogui Wan group (ZGW), Yougui Wan group (YGW), common prescription group (GTY), Yin prescription group (ZYY), Yang prescription group (BYY), progynova group (BJL) were prepared, and they simultaneously intervened the bone marrow mesenchymal stem cells (BMSCs). BMSCs proliferation was tested by the method of MTT, and BMSCs were identified by FCM (flow cytometry). The mRNA expression of Peroxisome Proliferator-Activated Receptor γ (PPARγ), lipoprotein (LPL) and fatty acid binding protein 4 (FABP4) were assayed using Q-PCR. Result All the serum containing Zuogui Wan and Yougui Wan and their disassembled prescriptions downregulated PPARγ, LPL and FABP4 mRNA. Especially, Yougui Wan remarkably downregulated PPARγ, LPL and FABP4 mRNA (P<0.05). Conclusion Zuogui Wan and Yougui Wan and their disassembled prescriptions can inhibit adipogenic differentiation .Yuogui Wan may have a higher level of inhibition on adipogenic differentiation than Zuogui Wan, yang prescription and common prescription.
