      目的 探讨染料木黄酮(genislein,Gen)对糖尿病大鼠股骨头生长板退化及骨质疏松的保护作用。方法 建立速发型链脲佐菌素(streptozotocin,STZ)糖尿病大鼠模型,随机分为正常组(CON)、糖尿病组(DM )、Gen干预组(Gen)。每天1次,灌胃给药(4.0 mg/kg),每组20只。15 w后光镜下作组织病理学分析及组织计量学测定;利用Van Gieson胶原纤维染色法观察胶原纤维沉积变化;ELISA测定血清蛋白聚糖和生化检测各组股骨头组织匀浆的醛糖还原酶(AR)活性。结果 CON组骨小梁粗厚、有完整的成骨细胞区。DM组骨小梁稀疏细薄、细胞核固缩;生长板厚度、生长板细胞柱密度均明显小于对照组(P<0.01);生长板钙化区胶原纤维明显增多;AR活性增大(P<0.01); 蛋白聚糖含量明显增加(P< 0.01)。Gen组骨小梁排列较紧密;生长板较DM组厚、生长板细胞柱密度大(P<0.01)。AR活性降低(P<0.01),蛋白聚糖下调(P<0.01)。结论 染料木黄酮可减轻糖尿病生长板的萎缩退化和骨质疏松变化,防止生长板过早闭合。

      Objective To investigate the mechanism of Genistein in preventing early closure of growth plate in diabetic rats. Methods The diabetic model (STZ) of rats was established. Rats were randomly divided into normal group (CON) , diabetic group (DM) and Gen group (4.0 mg/kgd),with 20 rats in each group. The changes of growth plate tissue structure were examined using microscopy, and changes in collagen fiber deposition were observed by using Van Gieson collagen fiber staining method in each group after 15 weeks. The content of decorin in serum was tested and the activity of AR was analyzed by biochemical detection at growth plate in all groups. Results The trabecular bone in group CON was thick and had complete osteoblasts area. Compared with the control group, the trabecular bone in group DM was sparse and thin,the nucleus was pyknotic and the thickness and cellular density of cell pillar in the growth plate were obviously lower at 15-week in the diabetic group (P<0. 05). The content of collagen in the calcified zone of the growth plate was higher in the diabetic group than that in the control group (P <0. 05),and the activity of AR and decorin were significantly increased (P <0. 01). The trabecular bone in Group Gen had arranged more closely. The thickness and cellular density of cell pillar were significantly increased (P <0. 01) , and the activity of AR and decorin were decreased in Group Gen (P <0. 01). Conclusion Gen could relieve atrophy degeneration of growth plate and the tendency of osteoporosis in diabetic rats. Gen could prevent early closure of growth plate to occur in diabetic rats.
