目的 观察葛根素对吡格列酮诱导的成骨细胞凋亡的影响。方法 MTT法检测不同浓度葛根素(0.01 -100μmol/L) 和吡格列酮(20μmol/L)作用于MC3T3-E1细胞24 h后细胞增殖活性影响。流式细胞仪测定葛根素和吡格列酮对MC3T3-E 细胞凋亡的影响。Western blot检测凋亡相关蛋白Caspase-3、Caspase-8、Bcl-2、Bax的表达情况。结果 1、与对照组相比,吡格列酮组的细胞增殖活性明显下降(P<0. 05 ),而0. 1 -10 (μmol/L葛根素组细胞增殖活性明显增强(P<0. 05 )。2、与吡格列酮组相比,加人0.1 -1μmol/L葛根素后细胞的增殖活性明显增强(P<0. 05 )。3、加人1μmol/L的葛根素后,吡格列酮导致的细胞凋亡率上升受到抑制(P<0. 05 )。4、吡格列酮处理后Caspase-3、Caspase-8、Bax蛋白的表达上调,Bcl-2蛋白的表达下调,而加入葛根素共同作用后,caspase-3蛋白表达下调,Bcl-2蛋白的表达上调。结论 葛根素可以抑制吡格列酮诱导的成骨细胞凋亡。 |

Objective To observe the effect of puerarin on osteoblasts apoptosis induced by pioglitazone. Methods Cell proliferation activity of MC3T3-E1 cells on different concentrations of puerarin(0.01 - 100μmol/L)and pioglitazone(20μmol/L) after 24 hours was detected by MTT method. Cell apoptosis of MC3T3-E1 cells on puerarin and pioglitazone was detected by flow cytometry. The expression of apoptosis related proteins caspase-3, caspase-8, Bcl-2 and Bax was detected by Western blot. Results 1, Compared with the control group, cell proliferation activity of pioglitazone group was obviously decreased (P < 0. 05) , but that of 0. 1 - 10μmol/L puerarin group was obviously increased ( P < 0. 05). 2. Compared with the pioglitazone group, cell proliferation activity was obviously increased after 0. 1 - 1μmol/L puerarin was added (P< 0. 05). 3. The apoptosis rate increased by pioglitazone was inhibited after 1μmol/L puerarin was added (P <0. 05). 4. Pioglitazone enhanced caspase-3, caspase-8 and Bax protein expression and decreased Bel - 2 protein expression. But caspase-3 protein expression was decreased and Bcl-2 protein expression was increased after puerarin was added. Conclusion Puerarin can inhibit the osteoblasts apoptosis induced by pioglitazone. |
