      目的 研究ERRα抑制剂(XCT-790)对沉默Wnt信号通路抑制因子Dickkopf1(DKK1)、骨硬化蛋白Sclerostin( Sost)重组腺病毒载体转染MG63细胞中OPG、CTGF、FGF2、TNFα的影响。方法 本实验将培养好的MG63细胞分为空白对照组、沉默DKK1组、沉默Sost组、沉默(DKK1 + Sost)组、XCT-790处理空载腺病毒组、XCT-790处理沉默DKK1组、XCT-790处理沉默Sost组、XCT-790处理沉默(DKK1 + Sost)组,根据组别不同分别用包装好的沉默DKK1、Sost腺病毒载体转染MG63细胞,应用 Western blot检测MG63细胞中OPG、CTGF、FGF2、TNFα蛋白的表达量。结果 (1 )与空白对照组对比,沉默DKK1、Sost、 DKK1 + Sost可以提高OPG、CTGF、FGF2表达量(P <0. 05),降低TNFα表达量(P <0. 05) ;XCT-790干预MG63细胞可降低OPG、CTGF、FGF2 表达量(P <0. 05),增加 TNFα表达量(P<0. 05) ;(2)与沉默 DKK1、Sost、DKKl + Sost 组比较,XCT-790 处理沉默DKKl、Sost、DKKl + Sost组可降低因沉默DKK1、Sost、DKKl + Sost而增加OPG、CTGF、FGF2的表达量的作用(P <0. 05 ), 增加因沉默DKK1、Sost、DKKl + Sost而降低TNFα的表达量(P <0. 05)。结论 XCT-790可以降低MG63细胞中OPG、CTGF、 FGF2表达量,升高TNF α表达量;还可以调节因沉默DKK1、Sost、DKK1 + Sost而改变的MG63细胞中OPG、CTGF、FGF2及 TNFα的表达量。

      Objective To investigate the effect of ERRα inhibitor (XCT-790) on silencing Dickkopfl (DKK1) , sclerostin (Sost) reconmbinant adenovirus vector, and the related proteins OPG, CTGF, FGF2, and TNFα in the transfection MG63 cells. Methods MG63 cells were divided into control group, silence DKK1 group, silence Sost group, silence (DKK1 + Sost) group, XCT-790 dispose no-load adenovirus group, XCT-790 dispose silence DKK1 group, XCT-790 dispose silence Sost group, and XCT-790 dispose silence (DKK1 + Sost) group. The silencing DKK1 or Sost reconmbinant adenovirus vector was transfected to MG63 cells, respectively. The expression levels of related proteins OPG, CTGF, FGF2, and TNFα were detected with Western blotting in MG63 cells. Results (1) Compared with control group, the expression levels of OPG, CTGF, FGF2 increased in silence DKK1, Sost, and DKK1 + Sost group ( P < 0. 05 ) , while the expression level of TNFα decreased ( P < 0, 05 ). The expression levels of OPG, CTGF, and FGF2 decreased (P <0. 05),while TNFα increased (P<0. 05) , when XCT-790 intervened MG63 cells. (2) Compared with silence DKK1, Sost, and DKK1 + Sost group, the increased expression levels of OPG, CTGF, FGF2 were impaired ( P < 0. 05) and the decreased expression level of TNFα increased (P < 0. 05) in XCT-790 dispose silence DKK1, Sost, and DKK1 + Sost groups. Conclusion XCT-790 decreases the expression levels of OPG, CTGF, and FGF2, increases the expression level of TNFα. It also regulates the changed expression levels of OPG, CTGF, FGF2, and TNFα in silencing DKK1,Sost , and DKK1 + Sost MG63 cells.
