      目的 探讨细胞因子IL-4, IL-6和TNF-α对脐带血来源的间充质干细胞成骨分化的作用。方法 取脐带血,以Percoll分离液离心分离细胞后进行培养,通过流式细胞仪检测间充质干细胞标志物CD29和CD44及造血干细胞标志物CD34;干细胞进行基础成骨分化培养后,以10ng/mL细胞因子IL-4、IL-6和TNF-α进一步诱导,检测ALP活力,以茜素红对细胞进行钙结节染色比较。结果 分离的细胞高表达CD29和CD44,基本不表达造血干细胞标志物CD34,为较纯的间充质干细胞;IL-4, IL-6诱导成骨9d后,ALP活力相比阴性对照组显著增加(P<0.01);IL-4、IL-6和TNF-α作用细胞18d时,钙结节的形成个数为(6.4±1.78)、(11.2±1.75)、(4.5±1.90),较阴性对照组(0.6±0.84)显著增加(P<0.01),其中IL-6作用后ALP活力增幅最大,钙结节形成的数目最多。结论 IL-4、 IL-6作用9d,可刺激间充质干细胞成骨分化,其中IL-6作用最为明显。

      Objective To explore the effects of interleukin-4, interleukin-6, and tumor necrosis factor-α (TNF-α) on the differentiation of osteoblasts by human umbilical cord blood mesenchymal stem cells. Methods Human umbilical cord blood mesenchymal stem cells (MSCs) were isolated with Percoll separating liquid and cultured. The cell surface antigens CD29, CD44, and CD34 were identified with flow cytometry. MSCs were induced with the basal osteogenic differentiation agent, and were further treated with 10ng/mL of IL-4, IL-6, or TNF-α. Alkaline phosphatase activity and Alizarin red staining were measured and compared. Results The selected cells expressed CD29 and CD44, but not CD34, suggesting that the cells were pure MSCs. Compared with negative control group, ALP activity was remarkably raised in cells treated with IL-4 or IL-6 for 9 days (P<0.01). After treatment of IL-4, IL-6, or TNF-α for 18 days, the number of calcium nodules were 6.4±1.78, 11.2±1.75, and 4.5±1.90, respectively, which were significantly higher than that in control cells (0.6±0.84, P<0.01). Among those, IL-6 strongly enhanced ALP activity and bone nodule formation. Conclusion IL-4 and IL-6 enhance osteogenic differentiation of human umbilical cord blood MSCs when applied for 9 days. It is the most obvious in IL-6 treatment.
