      目的 通过去卵巢结合蔗糖铁诱导家兔铁蓄积状态骨质疏松模型,以期为有关铁蓄积状态骨质疏松的实验研究提供动物模型基础。方法 将30只5月龄的新西兰雌性家兔随机分为对照组、去势组和铁蓄积组;每组分别于术前1天检测血清 1型前胶原氨基末端肽(P1NP),1型胶原羟基末端肽(CTX)及血清铁蛋白水平,并行腰椎骨密度测量;去势组和铁蓄积组手术切除双侧卵巢,对照组仅切开卵巢周围部分脂肪;1周后铁蓄积组耳缘静脉注射蔗糖铁,剂量为15 mg/kg,每周1次,共12周; 对照组和去势组以同样方式和频次给予等量生理盐水注射;所有组分别于术后4、8、12、16、20、24周检测血清P1NP,CTX及血清铁蛋白水平,并行腰椎骨密度测量。结果(1)所有家兔注射蔗糖铁后均未出现昏迷、抽搐、精神萎靡、呼吸急促等不良反应或致死的情况;(2)铁蓄积组血清铁蛋白水平在造模后8、12、16、20及24周均明显髙于对照组(P <0. 05);(3)铁蓄积组与 去势组血清P1NP,CTX水平在造模后12周、16周、20周及24周均明显髙于对照组(P <0. 05);铁蓄积组血清P1NP,CTX水平在造模后20周及24周均明显髙于去势组(P <0.05);(4)铁蓄积组与去势组腰椎骨密度丢失百分率均在造模后第24周超过 25%(33. 72%、25. 92%)。结论 去卵巢结合蔗糖铁可成功建立家兔铁蓄积状态骨质疏松模型。

      Objective Our study was purposed to evaluate the iron accumulation condition osteoporosis rabbit model which was established by ovariectomy combined with iron sucrose,and to develop a new method for inducing iron accumulation condition osteoporosis in rabbits. Methods 30 five-month old female New Zealand rabbits were randomly divided into three groups: control group (10 rabbits) ; ovariectomized group (OVX,10 rabbits) ; and iron accumulation group (IA,iron sucrose 15 mg/kg,10 rabbits). The bilateral ovaries of rabbits in the OVX and IA groups were removed,while those of the control group were not. The level of serum ferritin,P1NP,CTX and the lumbar spine bone mineral density (BMD) were observed at baseline and at 4,8,12, 16,20 and 24 weeks after the ovariectomy. Results (1) Iron accumulation rabbit model was induced by injecting iron sucrose with no adverse reactions or death. (2) Compared with the control group, serum ferritin in the IA group was significantly higher at 8, 12,16,20 and 24 weeks (P<0. 05). (3) Compared with the control group,P1NP and CTX in the IA and OVX groups were significantly higher at 12,16,20 and 24 weeks (P <0. 05),while P1NP and CTX in the IA group were significantly higher than those in the OVX group at 20 and 24 weeks (P <0. 05). (4) Compared with the control group,the decreased percentage of lumbar BMD in the IA and OVX groups were 33. 72% and 25. 92% . Conclusion Iron accumulation condition osteoporosis rabbit model can be successfully established by injecting iron sucrose combined with ovariectomy.
