      目的 制备并评价去势大鼠骨质疏松动物模型。方法 选取SD大鼠(雌性,2-3月龄)64只。随机分为:模型组(50 只)及正常组(14只),模型组采用摘除双侧卵巢结扎术制备骨质疏松雌性大鼠模型,俯卧位固定在大鼠固定板上,取背部切口,逐层切开,分离结扎输卵管,摘除双侧卵巢,依次缝合,碘伏消毒,预防感染(肌肉注射80万单位青霉素);正常组为对照组,自然饲养,不做任何处理。并采用大鼠阴道分泌物涂片法确定其动情周期以检测双侧卵巢摘除情况,自术后次日清晨开始,用无菌棉签获取各组大鼠阴道分泌物,涂片于载玻片,常规固定及风干后,进行瑞氏染色,连续5天。显微镜下观察阴道上皮角化细胞的形态变化。结果 模型组以白细胞为主,有核上皮细胞少见,呈连续性动情间期;正常组呈规律的动情周期变化:动情前期→动情期→动情后期→动情间期。结论 雌性大鼠去双侧卵巢可有效制备骨质疏松模型;采取观察阴道上皮角化细胞的检测方法是证明去势骨质疏松模型成功的标准。

      Objective To prepare and evaluate the animal model of osteoporosis in ovariectomized rats. Methods Sixty-four SD rats (female,2-3 months old) were selected,and randomly divided into model group (50) and normal group (14). The model group had the excision of bilateral ovary and ligation to prepare female osteoporosis rat model. Rats was fixed on the rat fixed plate in the prone position,then the ovariectomy was preceded by dorsal incision,incision and separation of tissues layer by layer,tubal ligation and ovariectomy,followed by suture,iodophor disinfection and prevention of infection (intramuscular injection of 800 thousand units of penicillin). The normal control group received natural feeding,without any treatment. In order to detect and determine the estrous cycle of bilateral ovariectomy rats by vaginal smear method ,from the morning after the day of surgery,rat vaginal secretions were harvested using sterile cotton,smeared on slides,which were then conventional fixed and dried by Wright ’ s stain for 5 consecutive days. Morphological changes of vaginal epithelial keratinocytes were observed under microscope. Results The model group mainly consisted of white blood cells,nuclear epithelial cells were rare,and they showed continuous diestrus. The normal group showed regular changes of estrous cycle: proestrus,estrus,metoestrus and diestrus. Conclusion The model of osteoporosis can be effectively prepared by bilateral ovariectomy in female rats, and the method of observing vaginal epithelial keratinocytes is the standard to prove the successful establishment of the ovariectomized osteoporosis model.
