      目的 探讨RhoA/ROCK信号通路在骨质疏松大鼠骨髓间充质干细胞(bone mesenchymal stem cells,BMSCs)成骨分化中的作用。方法 实验细胞分为两组,A组为骨质疏松大鼠BMSCs(去卵巢法制备骨质疏松模型),B组为3月龄大鼠BMSCs,通过BMSCs培养观察细胞生长情况、矿化结果,并用Western blot及PCR方法检测RhoA、ROCKl在两组细胞中的表达情况。结果 3月龄组细胞生长较为活跃,矿化能力强,Western blot显示骨质疏松组细胞内RhoA、ROCKl蛋白表达较3月龄组明显降低,差异有统计学意义(P <0.05),PCR结果显示骨质疏松组细胞内RhoA、ROCKl mRNA含量较骨质疏松组明显降低(P < 0.05)。结论 RhoA/ROCK信号通路在骨质疏松BMSCs中表达降低,这可能是骨质疏松发病的重要原因之一。

      Objective To explore the role of RhoA/ROCK signaling pathway in the osteoblast differentiation of bone mesenchymal stem cells ( BMSCs) in osteoporotic rats. Methods BMSCs were divided into two groups,group A from osteoporotic rats and group B from 3-month-old rats. The growth morphology and mineralization of BMSCs were observed. The expression of RhoA and ROCK protein was examined with Western blotting and the expression of RhoA and ROCK mRNA was examined with PCR. Results The 3-month BMSCs had the more vigorous growth than the osteoporosis BMSCs,and the mineralization was more in 3-month BMSCs. Western blotting result showed that the expression of RhoA and ROCK protein in osteoporosis group was significant lower than that in the 3-month BMSCs (P <0. 05). The expression of RhoA and ROCK mRNA in the osteoporosis BMSCs was significant lower than that in 3 -month group (P <0. 05). Conclusion The expression of RhoA/ ROCK signaling pathway is lower in osteoporosis BMSCs,which may be one of the important cause of osteoporosis.
