补肾健脾活血方含药血清对UMR106细胞成骨分化和骨形成相关蛋白的影响

      目的 观察补肾健脾活血方含药血清对UMR106细胞成骨分化和骨形成相关蛋白的影响。方法 将实验动物随机分成空白血清组、低剂量血清组、中剂量血清组、高剂量血清组、阳性对照组,分别予以灌胃制备含药血清,并对其细胞进行加药培养,测定UMR106 细胞培养72h后的检测增殖、ALP 活性以及各组细胞OPN、RUNX2、BMP-2蛋白表达情况。结果 与空白血清组比较,低、中、高剂量血清组和阳性对照组加药干预72h后,细胞吸光度值均较低(P<0.05),而细胞ALP活性均较高(P<0.05),且随着中药血清水平升高,ALP活性提高;与空白血清组相比,经中药含药血清培养后UMR106细胞OPN、RUNX2、BMP-2均显著上调(P<0.05),且呈浓度梯度性变化,各中药含药血清组以高剂量组表达最为明显。结论 表明补肾健脾活血方(骨康方)含药血清有促进UMR 106细胞成骨分化和调节骨形成相关蛋白;且随着补肾健脾活血方(骨康方)水平的增高,成骨分化更明显。

      Objective To obverse the effect of nourishing kidney, invigorating spleen, and promoting blood flow medicine (Gukang) containing serum on osteogenic differentiation and bone formation-related proteins in UMR106 Cells. Methods The experimental animals were randomly divided into blank serum group, low-dose serum group, medium-dose serum group, high-dose serum group, and positive control group. Gukang with different doses was intragastricly administered to prepare the drug containing serum, and the cells were cultured with the serum. The proliferation of UMR106 cells, the activity of ALP, and the expression of OPN, RUNX2, and BMP-2 proteins in UMR106 cells were measured after 72 hours of culture. Results Compared with the blank serum group, the median absorbance was lower in the medium- and high-dose serum group and the positive control group in 72 hours (P <0.05), but ALP activity was higher (P <0.05). Compared with the blank serum group, the protein levels of OPN, RUNX2, and BMP-2 in UMR106 cells increased significantly in a dose-dependent manner (P<0.05). Conclusion Gukang containing serum promotes the osteogenic differentiation of UMR 106 cells and up-regulates bone morphogenetic proteins in a dose-dependent manner.
