      目的 探讨去卵巢对大鼠颌骨成骨细胞增殖与成骨分化能力的影响。 方法 建立去卵巢大鼠骨质疏松模型,分离培养假手术组和去卵巢组颌骨成骨细胞并进行形态学观察。采用CCK-8法检测假手术组和去卵巢组大鼠颌骨成骨细胞的增殖能力差异。通过碱性磷酸酶染色与活性测定、矿化结节茜素红染色与定量、成骨相关基因及蛋白表达检测,探讨去卵巢对大鼠颌骨成骨细胞增殖及成骨分化能力的影响。 结果 假手术组和去卵巢组大鼠均可成功分离培养颌骨成骨细胞,两组细胞均可贴壁生长,细胞形态未见显著差异。与假手术组相比,去卵巢大鼠颌骨成骨细胞的增殖能力,碱性磷酸酶染色与活性均显著增高;但去卵巢大鼠颌骨成骨细胞的Runx2和OC在mRNA与蛋白水平的表达上及体外矿化结节形成能力方面皆显著低于假手术组。结论 去卵巢可影响大鼠颌骨成骨细胞的增殖与成骨分化能力,研究结果将为探讨颌面部骨组织骨质疏松的发病机制与防治提供实验依据。

      Objective To explore the effect of ovariectomy on proliferation and osteogenic differentiation of osteoblasts from rat mandibles. Methods Osteoporotic rat model was established by ovariectomy. Osteoblasts were isolated and cultured from mandibles of ovariectomized group (ovx group) and sham-operated group (sham group). The morphological appearance of osteoblasts from rat mandibles was observed with a microscope. To explore the influence of ovariectomy, the proliferation and osteogenic differentiation of osteoblasts from ovx group and sham group were evaluated with CCK-8 assay, ALP staining and activity, Alizarin red S staining, quantification real-time PCR and Western blotting. Results Mandibular osteoblasts were successfully isolated and cultured from both sham group and ovx group and there was no significant difference in the morphological appearance. Comparing to sham group, the proliferation of osteoblasts of ovx group significantly elevated. The expression of ALP in osteoblasts in ovx group was significantly higher than that in sham group. However, the mRNA and protein expressions of Runx2 and OC and calcium nodule formation of osteoblasts in ovx group were lower. Conclusion Ovariectomy affects the proliferation and osteogenic differentiation of osteoblasts from rat mandibles. These results provide an experimental evidence for discussing the pathogenesis and treatment of osteoporosis of craniofacial bone.
