      目的 研究鹿角补骨颗粒对大鼠骨代谢的影响及急性毒性检测,确定临床用药的疗效及安全性。方法 大鼠骨代谢实验:将大鼠随机分为低剂量组、高剂量组、空白组,给予低剂量组、高剂量组大鼠鹿角补骨颗粒连续灌喂6 个月,空白组灌喂蒸馏水6个月,给药结束后测定三组大鼠血清Ⅰ型前胶原氨基末端(type I collagen N terminal peptide ,PINP) 、骨钙素(bone glaprotein,BGP)、Ⅰ型胶原交联羧基末端肽(type I collagen carboxy-terminal peptide,CTX)含量,并与空白对照组比较。急性毒性实验:采用最大耐受实验法,以116.4 g/kg剂量给予小鼠灌喂鹿角补骨颗粒,连续观察7 d。结果 骨代谢结果:鹿角补骨颗粒能够提高大鼠血清PINP、BGP水平,且随灌喂剂量增加呈上升趋势;高剂量组大鼠CTX水平低于低剂量组,均高于空白组。急性毒性实验:动物小鼠活动正常,未见中毒及死亡症状。结论 鹿角补骨颗粒能够改善大鼠骨代谢且对实验小鼠无明显急性毒副作用。

      Objective To study the effect of Lujiaobugu Granule on bone metabolism in rats and its acute toxicity, and to determine the efficacy and safety of its clinical application. Methods Rat bone metabolism test: the rats were randomly divided into low dose group, high dose group and blank group. Rats in low dose group and high dose group were given Lujiaobugu Granule gavage for 6 months continuously, and the blank group was fed with distilled water for 6 months. After the administration, the contents of serum type Ⅰ procollagen (PINP), osteocalcin (BGP) and type Ⅰ collagen cross-linked carboxyl terminal peptide (CTX) were measured and compared with the blank control group. Acute toxicity test: the maximum tolerated experimental method was used to gavage mice with antlers at a dose of 116.4 g/kg, and continuously observed for 7 days. Results Bone metabolism results: Lujiaobugu Granule could increase the level of serum PINP and BGP in rats, and the increase was greater with increased dose of gavage. The level of CTX in the high dose group was lower than that in the low dose group. Acute toxicity test: mice were normal, and no symptoms of poisoning and death were observed. Conclusion Lujiaobugu Granule could improve bone metabolism in rats and has no obvious acute side effects on experimental mice.
