      目的 探讨北京人群乳腺癌雌激素调控基因1(GREB1)基因多态性与老年骨质疏松性骨折的相关性。方法 本研究共纳入226例老年患者,分为骨折组(骨质疏松性骨折,n=100)、对照组(无骨质疏松、无骨折,n=126)。采用双能X线骨密度仪测量髋部和腰椎骨密度值。用Snapshot技术检测患者rs10929757、rs5020877位点多态性。比较两组总体及亚组在GREB1各位点的等位基因频率、基因型频率分布差异。比较GERB1各位点不同基因型患者的骨密度值差异。结果 两组总体在GREB1基因rs10929757、rs5020877位点的基因频率和基因型频率分布均无明显差异(P>0.05)。两组男性患者在rs10929757位点的C等位基因频率分别为骨折组36.5%、对照组52.2%,差异有统计学意义(P<0.05);两组男性患者在rs5020877位点的基因频率和基因型频率分布均无明显差异(P>0.05)。两组女性患者在rs10929757位点和rs5020877位点的基因频率和基因型频率分布无明显差异(P>0.05)。在rs10929757位点,总体、男性和女性患者中不同基因型患者骨密度值比较无明显差异(P>0.05)。在rs5020877位点,总体、男性和女性患者中不同基因型患者骨密度值比较无明显差异(P>0.05)。结论 rs10929757位点多态性与老年男性椎体骨质疏松性骨折存在相关性;未发现rs5020877位点多态性与老年椎体骨质疏松性骨折相关。

      Objective To investigate the relationship between the growth regulation by estrogen in breast cancer 1 (GREB1) gene polymorphism and the incidence of osteoporotic fracture in Han Beijing senile population. Methods A total of 226 elderly patients were recruited and divided into fracture group (with osteoporotic fracture, n=100) and control group (without osteoporosis and fracture, n=126). The bone mineral density (BMD) of hip and lumbar spine were measured using a dual-energy X-ray bone densitometer. The genotype of GREB1 gene rs10929757 and rs5020877 locus were examined by Snapshot technique. Allele frequency and genotype frequency at both locus were compared. The patients’ BMD values of different genotype at both locus were compared. Results Between the two groups, there were no statistical differences in the distribution of allele frequency and genotype frequency at both locus (P>0.05). Between the two groups of males, the C allele frequencies at rs10929757 locus were 36.5% in the fracture group and 52.2% in the control group, respectively, with statistical differences (P<0.05); there were no statistical differences in the allele frequencies of rs5020877 locus (P>0.05). Between the two groups of females, there were no statistical differences in the allele frequencies of both locus (P>0.05). When comparing patients’ BMD values, at rs10929757 locus, there were no statistical differences among different genotypes in the whole sample, males and females; at rs5020877 locus, there were no statistical differences among different genotypes in the whole sample, males, and females. Conclusion The polymorphism of rs10929757 is associated with vertebral osteoporotic fracture in the Beijing Han senile male population. The polymorphism of rs5020877 is not associated with vertebral osteoporotic fracture in the Beijing Han senile population.
