Objective To investigate the effects of Xianlinggubao Capsule on the expression levels of Estrogen Receptor α (ERα), histone demethylase (JMJD2B), and Runt-related transcription factor 2 (Runx2) and Alkaline Phosphatase (ALP). Methods Forty SD female rats were divided into four groups according to the random number table: blank control group, negative control group, experimental group and positive control group, with 10 rats in each group. Rats in the blank control group retained bilateral ovaries and only excised paraesophageal fat. Rats in the other three groups were subjected to surgical resection of bilateral ovaries to prepare an osteoporotic model. One week after surgery, the experimental group was given Xianlinggubao Capsules at a ratio of 30.85 mg/100 g, the positive control group was given FuMeiJia at a ratio of 11.7 mg/100 g, and the blank control group and the negative control group were fed with the same volume of distilled water. After 3 months of continuous medication, RT-PCR was used to detect the mRNA expression of ERα, JMJD2B, Runx2 and ALP in rat femur. Western Blot was used to detect the expression of ERα and JMJD2B protein. Results Compared with the blank control group, the expression of ERα, JMJD2B protein and its mRNA, Runx2 and ALP in the femur tissue of the experimental group decreased significantly, and there were statistically significant differences between the two groups (P<0.05). Compared with the negative control group, the expression of ERα, JMJD2B protein and its mRNA, Runx2 and ALP in the experimental group increased significantly (P<0.05). Compared with the positive control group, the expression of ERα, JMJD2B protein and its mRNA in the experimental group increased significantly, with statistically significant differences between the two groups (P<0.05), while the expression level of Runx2 decreased significantly (P<0.05).Conclusion The results of this experiment found that Xianlinggubao Capsule could increase the expression of ERα, JMJD2B, Runx2 and ALP. |
