      目的 探讨补骨生髓方对去势骨质疏松大鼠Smad/ERK信号通路的影响。方法 通过采取去性腺法(摘除睾丸法)建立骨质疏松症大鼠实验模型,造模成功后按照实验方案对大鼠进行灌胃治疗,2个月后处死各组大鼠,HE常规染色,并通过免疫组织化学染色的方法检测正常对照组及各中药干预组去势大鼠ERK、p-ERK、Smad4蛋白的表达。结果 与模型组比较,补骨生髓方高剂量组与中剂量组ERK蛋白的表达差异有统计学意义(P<0.05),而低剂量组与模型组表达差异无统计学意义(P>0.05);与模型组比较,补骨生髓方高剂量组与中剂量组p-ERK蛋白的表达差异有统计学意义(P<0.05),而低剂量组与模型组表达差异无统计学意义(P>0.05);与模型组比较,补骨生髓方高剂量组与中剂量组Smad4蛋白表达差异有统计学意义(P<0.05),而低剂量组与模型组比较差异无统计学意义(P>0.05)。结论 补骨生髓方在一定程度上能够上调ERK、p-ERK、Smad4蛋白的表达水平,其可能是通过调控ERK蛋白磷酸化介导的Smad/ERK信号通路来发挥作用;ERK、Smad4可能是今后治疗骨质疏松的潜在作用靶点,但仍需要进一步开展更深入的研究。
英文摘要:
      Objective To investigate the effect of Bu Gu Sheng Sui decoction on Smad/ERK signaling pathway in ovariectomized osteoporosis rats. Methods The experimental model of osteoporosis rat was established by adopting the gonadal method. After successful modeling, the rats were treated with gavage according to the experimental protocol. After 2 months, the rats in each group were sacrificed, the routine HE staining was performed and the immunohistochemical staining was used to detect the expressions of ERK, p-ERK and Smad4 proteins in ovariectomized rats after normal control group and Chinese medicine intervention. Results Compared with the model group, the expression of ERK protein in the high-dose group and the middle-dose group was significantly different (P<0.05), but the difference between the low-dose group and the model group was not statistically significant (P>0.05). Compared with the model group, the expression of p-ERK protein in the high-dose group and the middle-dose group was significantly different (P<0.05), but the difference between the low-dose group and the model group was not statistically significant (P>0.05). Compared with the model group, there was a statistically significant difference in the expression of Smad4 protein in the high-dose group and the middle-dose group (P<0.05), but there was no significant difference between the low-dose group and the model group (P>0.05). Conclusion Bu Gu Sheng Sui decoction may affect the expression levels of ERK, p-ERK and Smad4 proteins, which is likely to play a role in regulating the Smad/ERK signaling pathway mediated by phosphorylation of ERK protein. ERK and Smad4 may be important targets for treating osteoporosis, but further research is needed.
