左、右归丸通过调节AMPK/mTOR通路抑制大鼠股骨成脂化的实验研究

      目的 基于AMPK/mTOR信号通路初步探讨左、右归丸对绝经后骨质疏松(postmenopausal osteoporsis,PMOP)大鼠成脂分化的调节机制。方法 将60只雌性SD大鼠随机分为空白组(KB)、假手术组(SHAM)、模型组(OVX)、左归丸组(ZGW)、右归丸组(YGW)、补佳乐组(BJL)。除KB、SHAM组外,其余大鼠均摘除双侧卵巢,SHAM组大鼠摘除双侧卵巢周围等量脂肪,灌胃12周。应用数字化全身骨密度仪检测大鼠右侧股骨骨密度(bone mineral density,BMD),应用实时荧光定量PCR法检测大鼠右侧股骨PPARγ、AMPK、mTORC1 mRNA的表达,采用Western blot法检测大鼠右侧股骨PPARγ、C/EBPα、C/EBPβ蛋白的表达以及AMPKα、mTOR蛋白磷酸化水平。结果 左、右归丸能明显升高PMOP大鼠右侧股骨BMD(P<0.01),降低大鼠股骨成脂相关mRNA与蛋白的表达(P<0.01或P<0.05),降低AMPK mRNA的表达与蛋白磷酸化水平(P<0.01),升高mTORC1 mRNA的表达与蛋白磷酸化水平(P<0.01或P<0.05);与ZGW组相比,YGW组PMOP大鼠右侧股骨BMD没有明显差异(P>0.05),股骨成脂分化相关mRNA与蛋白的表达明显升高(P<0.01),且AMPK mRNA的表达明显升高(P<0.01),mTORC1 mRNA的表达与蛋白磷酸化水平降低(P<0.01或P<0.05)。结论 左、右归丸通过AMPK/mTOR通路改善了PMOP大鼠股骨成脂分化过度,其作用机制可能与调节AMPK、mTOR mRNA的表达与蛋白磷酸化水平有关。

      Objective This study was based on AMPK/mTOR signaling pathway, preliminarily investigated the regulation mechanism of Zuo/You gui pills on lipogenic differentiation in(postmenopausal osteoporsis,PMOP)rats. Methods Divided sixty SD female rats into six groups randomly: blank group (KB), sham operation group (SHAM), model group (OVX), Zuo gui pills group (ZGW), You gui pills group (YGW) and progynova group (BJL). Except for KB and SHAM group, all the other rats were ovariectomized. In SHAM group, the same amount of fat around the ovaries was removed and gavage for 12 weeks.Used digital whole-body bone densitometer to detect the BMD of the right femur of rats, used real-time fluorescence quantitative PCR to detect the expression of PPARγ, AMPK, mTORC1 mRNA in the right femur of rats, and used Western blot to detect the expression of PPARγ,C/EBPα and C/EBPβ protein and the phosphorylation level of AMPKαand mTOR protein in right femur of rats.Results Zuo/You gui pills significantly increased PMOP rats right femur BMD(P<0.01);decreased the expression of lipogenic mRNA and protein of femur(P<0.01 or P<0.05),decreased AMPK mRNA expression and protein phosphorylation level(P<0.01) ,and increased mTORC1 mRNA expression and protein phosphorylation level(P<0.01 or P<0.05).Compared with ZGW group,PMOP rats in the YGW group showed no significant difference in right femur BMD(P>0.05),significantly increased the expressions of adipogenic differentiation related mRNA and protein of femur(P<0.01),significantly increased the expressions of AMPK mRNA(P<0.01),and decreased the expressions of mTORC1 mRNA and protein phosphorylation(P<0.01 or P<0.05). Conclusion Zuo/You gui pills can improve the over differentiation of femur in PMOP rats through AMPK / mTOR pathway. The mechanism may be related to the regulation of AMPK, mTOR mRNA expression and protein phosphorylation level.
