      目的 观察聚丙交酯-乙交酯(PLGA)微球搭载鹿茸多肽(velvet antler polypeptides, VAP)对去卵巢骨质疏松模型大鼠的保护作用,并探讨相关作用机制。方法 摘除大鼠双侧卵巢复制骨质疏松动物模型。48只大鼠随机分为假手术组、模型对照组、阿仑膦酸钠组、PLGA搭载VAP的低、中、高剂量组,每组8只,连续给药3个月。运用生化分析仪检测各组大鼠血清中钙离子(Ca2+)、酸性磷酸酶(acid phosphatase, ACP)、碱性磷酸酶(alkaline phosphatase, ALP)、骨钙素(osteocalcin, OC)水平,通过骨密度仪检测各组大鼠股骨骨密度(bone mineral density, BMD),HE染色法检测各组大鼠骨皮质平均厚度(bone cortical thickness, BCT)和骨小梁面积(femur trabecular area, FTA),Western blot检测各组大鼠股骨Runx2(runt-related transcription factor 2)和骨形成蛋白2(bone morphogenetic protein 2, BMP-2)表达水平。结果 PLGA微球搭载VAP可明显降低骨质疏松大鼠血清ACP、ALP、和OC含量,提高血清Ca2+、股骨BMD、BCT及FTA水平,同时提高大鼠股骨Runx2和BMP-2蛋白表达水平(P<0.05或P<0.01)。 结论 PLGA微球搭载VAP对去卵巢大鼠骨质疏松症有明显的保护作用,其作用机制可能与提高Runx2和BMP-2蛋白表达水平有关。

      Objective To observe the protective effects of PLGA microspheres loaded VAP on ovariectomized osteoporosis rats and to explore its related mechanism. Methods The osteoporosis models were made by removing bilateral ovariectomy of rats. 48 rats were randomly divided into sham group, model control group, alendronate group, PLGA loaded VAP high-dose group,middle-dose group and low-dose group(n=8) and continued giving medicine for 3 months. The levels of Ca2+, acid phosphatase (ACP), alkaline phosphatase(ALP), osteocalcin(OC) of rats in serum were detected by biochemical analyzer. Bone mineral density(BMD) of rats were detected by bone density detector. Bone Cortical Thickness(BCT) and femur trabecular area(FTA) of rats were detected by HE staining. The protein expression levels of runt-related transcription factor 2 (Runx2) and bone morphogenetic protein 2 (BMP-2) in femur of rats were detected by western blot. Results PLGA microspheres loaded with VAP could significantly reduce the content of serum ACP, ALP, and OC in rats with osteoporosis, increase the levels of serum Ca2+, BMD in femur, BCT and FTA. Meanwhile,it also could increase the expression levels of Runx2 and bmp-2 proteins in femur of rats with osteoporosis. Conclusion PLGA loaded VAP have significant protective effects on ovariectomized osteoporosis rats and its mechanism may be related to the increase of Runx2 and BMP-2 protein expression levels.
