Objective To observe the effects of Peiben Gushu Decoction on bone mineral density, bone biomechanics and bone metabolism in ovariectomized osteoporosis model rats. Methods 42 SD rats were randomly divided into sham operation group (SHAM), model control group (OVX), Chinese medicine low-dose group (D), Chinese medicine middle-dose group (Z), Chinese medicine high-dose group (G) and western medicine group (X), the osteoporosis model was established by removing bilateral ovaries of rats, and the sham group only removed about 1g of adipose tissue around the ovaries. Drug intervention was performed 4 weeks after modeling, and each group was continuously administered for 8 weeks. The uterus index was calculated, and the femoral bone density and femoral biomechanical elastic load, ultimate load, elastic modulus and other indicators were determined. The serum bone metabolism indexes TRACP5b, sRANKL, OPG and oxidative stress indicators MDA and AGEs were determined by ELISA. Results Compared with the SHAM operation group, the uterus index, BMD of the proximal femur and BMD, elastic load and elastic modulus of the femur were significantly decreased (P<0.05). The serum MDA and AGEs levels in the model group were significantly increased (P<0.01), the serum TRACP5b content increased significantly (P<0.01), while the OPG, OPG/sRANKL ratio decreased (P<0.01); compared with the model group: the uterus index and bone strength of each treatment group improved. The BMD of Peiben Gushu Decoction, high dose group and Alendronate group were significantly increased (P<0.05). The content of TRACP5b and OPG was decreased (P<0.05), The content of RANKL and OPG/sRANKL ratio were increased (P<0.05). The serum levels of MDA and AGEs in the high-dose group were significantly lower (P<0.05). Conclusion Peiben Gushu Decoction can increase bone strength, inhibit osteoclast formation through OPG/RANKL/RANK pathway, slow bone matrix uptake, and anti-oxidative stress mechanism may also be involved. The specific mechanism remains to be explored. |
