      目的 确定硫辛酸(ALA)对老年大鼠模型的保护作用,并探讨其作用机制。方法 将大鼠随机分为以下各组:青年对照组(Con),老年组(Old),老年+硫辛酸组(ALA)。适应性饲养7 d后给予ALA治疗12周。通过ELISA检查分析骨代谢指标。蛋白质印迹分析以评估NF-κB、Caspase-3和PARP蛋白表达。结果 ALA能显著改善老年大鼠的骨密度和骨量。老年大鼠在ALA给药治疗后可显著降低TRACP-5b和β-CTX水平(P<0.01);显著增加BLAP和OPG水平(P<0.01)。用ALA治疗可显著降低老年大鼠的NF-κB、Caspase-3和PARP蛋白表达,抑制NF-κB通路传导。结论 ALA可以通过抑制老年大鼠NF-κB信号通路对骨量流失和骨密度降低起到保护作用。

      Objective The purpose of this study was to determine the protective effect of lipoic acid (ALA) on aged rat models and to explore its mechanism of action. Methods The rats were randomly divided into the following groups: the young rats control group (Con), the old rats group (Old), and the old rat + lipoic acid group (ALA). After adaptive feeding for 7 days, ALA was given for 12 weeks. ELISA analysis is used to analyze the bone metabolism indicators. Western blot analysis was performed to assess NF-κB, Caspase-3 and PARP protein expression. Results The results of this study indicate that ALA can significantly improve bone density and bone mass in aged rats. Elderly rats can significantly reduce the levels of TRACP-5b and β-CTX after ALA administration (P <0.01), and significantly increase the levels of BLAP and OPG (P <0.01). Treatment with ALA can significantly reduce the expression of NF-κB, Caspase-3 and PARP in elderly rats, and inhibit NF-κB pathway conduction. Conclusion This study suggests that ALA can protect the bone mass by inhibiting NF-κB signaling pathway in aged rats.
