      目的 探讨GYY4137对去卵巢诱导的骨质疏松大鼠的影响。方法 将80只大鼠随机分为4组,采用雌性SD大鼠双侧卵巢全切除术(OVX)或假手术(Sham),建立骨质疏松模型,通过骨密度的检测确认模型建立成功,随后OVX大鼠给予腹腔注射GYY4137和阿仑膦酸钠治疗。测定血浆硫化氢(H2S)、血清碱性磷酸酶(ALP)活性、骨钙素(OCN)、降钙素、甲状旁腺激素和瘦素水平。采用双能X线骨密度仪测定左侧股骨密度(bone mineral density,BMD)。采用股骨三点弯曲试验获得股骨骨折的最大应力。结果 成功建立OVX骨质疏松模型。OVX-GYY组在观察期内注射GYY4137显著提高血浆H2S水平(P<0.05)。12周时OVX-GYY组大鼠股骨密度增加(P<0.05),OVX组股骨BMD值明显降低(P<0.05)。双侧卵巢切除可引起大鼠生化骨代谢和血浆激素水平的改变(P均<0.05)。卵巢切除还可以降低血钙、血磷和降钙素,增加甲状旁腺激素和瘦素。阿仑膦酸钠组和GYY4137组则显著改善上述指标(P均<0.05)。结论 GYY4137对去卵巢大鼠骨质疏松有积极的治疗作用。

      Objective The study aimed to explore the effect of GYY4137 on ovariectomy-induced osteoporosis rats. Methods Eighty rats were randomly divided into 4 groups. Female SD rats were treated with bilateral ovarian resection (OVX) or sham operation (Sham) to establish an osteoporosis model. Bone mineral density (BMD)was examined to confirm the establishment of the model. OVX rats received intraperitoneal injection of GYY4137 and alendronate treatment. The serum level of H2S, alkaline phosphatase (ALP) activity, and the levels of osteocalcin (OCN), calcitonin, parathyroid hormone, and leptin were determined. BMD of the femur was measured using a dual-energy X-ray absorptiometry. The maximal stress of femoral fracture was obtained through a three-point bending test. Results The OVX osteoporosis model was successfully established. The serum level of H2S increased after GYY4137 during the observation period in OVX-GYY group (P<0.05). At 12 weeks, BMD value of the femur in the OVX-GYY group increased (P<0.05), but it decreased in OVX-vehicle group (P<0.05). The bone metabolism and hormone levels changed after bilateral ovariectomy in rats (all P<0.05). The levels of calcium, phosphate, and calcitonin reduced, but the levels of parathyroid hormone and leptin increased in rats after ovariectomy. The indicators were improved in the groups with alendronate sodium or GYY4137 treatment (all P<0.05). Conclusion GYY4137 plays a positive role in the treatment of osteoporosis in ovariectomized rats.
