      目的 探讨丹参注射液对小鼠骨质疏松合并动脉粥样硬化的影响。方法 选用36只C57BL/6小鼠,随机分为对照组、模型组、低剂量丹参组(0.5 mL/kg)、中剂量丹参组(3.0 mL/kg)、高剂量丹参组(8.0 mL/kg)和辛伐他汀组(0.6 mg/kg),每组6只。采用卵巢摘除并高脂饲料喂养建立骨质疏松合并动脉粥样硬化症模型,给予相应药物进行干预。Elisa试剂盒检测血清中骨碱性磷酸酶(BALP)、骨钙素(OC)、抗酒石酸酸性磷酸酶5(TRAP5)、氧化性低密度脂蛋白(ox-LDL)水平;全自动生化分析仪测定血清中三酰甘油(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)水平;HE染色观察主动脉组织病理变化以及骨组织形态学变化。结果 与模型组比较,丹参各剂量组和辛伐他汀组小鼠股骨组织和主动脉组织病变均有不同程度减轻,且血清中BALP、OC、TRAP5、TG、TC、LDL-C、ox-LDL水平降低,HDL-C水平升高。结论 丹参注射液可以通过调节骨代谢平衡和血脂水平,对小鼠骨质疏松合并动脉粥样硬化症具有一定的缓解作用。

      Objective To investigate the effect of Danshen injection on osteoporosis combined with atherosclerosis in mice. Methods Thirty six C57BL/6 mice were randomly divided into control group, model group, low dose Danshen group (0.5 mL/kg), medium dose Danshen group (3.0 mL/kg), high dose Danshen group (8.0 mL/kg), and simvastatin group (0.6 mg/kg), with 6 mice in each group. The model of osteoporosis combined with atherosclerosis was established by using ovariectomy and high-fat feed. The corresponding drugs were given for intervention. Serum levels of bone alkaline phosphatase (BALP), osteocalcin (OC), tartrate-resistant acid phosphatase 5 (TRAP5), and oxidized low-density lipoprotein (ox-LDL) were measured using ELISA kits. Serum levels of triglyceride (TG), total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C), and high-density lipoprotein cholesterol (HDL-C) were measured with a full-automatic biochemical analyzer. HE staining was used to observe pathological changes of the aortic tissue and morphological changes of the bone tissue. Results Compared with those in the model group, the pathological changes of the femur and aorta in Danshen and simvastatin group were alleviated in varying degrees, the serum levels of BALP, OC, trap5, TG, TC, LDL-C and ox LDL decreased, and the levels of HDL-C increased. Conclusion Danshen injection alleviates osteoporosis combined with atherosclerosis in mice by regulating the balance of bone metabolism and the level of blood lipid.
