Objective To investigate the effect of Danshen injection on osteoporosis combined with atherosclerosis in mice. Methods Thirty six C57BL/6 mice were randomly divided into control group, model group, low dose Danshen group (0.5 mL/kg), medium dose Danshen group (3.0 mL/kg), high dose Danshen group (8.0 mL/kg), and simvastatin group (0.6 mg/kg), with 6 mice in each group. The model of osteoporosis combined with atherosclerosis was established by using ovariectomy and high-fat feed. The corresponding drugs were given for intervention. Serum levels of bone alkaline phosphatase (BALP), osteocalcin (OC), tartrate-resistant acid phosphatase 5 (TRAP5), and oxidized low-density lipoprotein (ox-LDL) were measured using ELISA kits. Serum levels of triglyceride (TG), total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C), and high-density lipoprotein cholesterol (HDL-C) were measured with a full-automatic biochemical analyzer. HE staining was used to observe pathological changes of the aortic tissue and morphological changes of the bone tissue. Results Compared with those in the model group, the pathological changes of the femur and aorta in Danshen and simvastatin group were alleviated in varying degrees, the serum levels of BALP, OC, trap5, TG, TC, LDL-C and ox LDL decreased, and the levels of HDL-C increased. Conclusion Danshen injection alleviates osteoporosis combined with atherosclerosis in mice by regulating the balance of bone metabolism and the level of blood lipid. |
