目的 观察去卵巢骨质疏松症大鼠骨组织Leptin启动子甲基化,探索补肾健骨中药复方防治绝经后骨质疏松症(postmenopausal osteoporosis,PMOP)的分子机制。方法 切除雌性Wistar大鼠双侧卵巢建立PMOP模型,实验大鼠分为4组:正常组、模型组、补肾健骨中药复方组、仙灵骨葆阳性对照组。给药14周后,X射线骨密度测定仪检测骨密度、显微镜观察股骨头石蜡切片HE染色显微形态结构、质谱法检测Leptin启动子甲基化。结果 ①第1~6腰椎骨密度,模型组与正常组相比显著降低(P<0.01);补肾健骨中药复方组、仙灵骨葆阳性对照组与模型组相比显著升高(P<0.05)。②模型组与正常组相比,股骨头骨皮质骨板变薄,骨松质骨小梁明显变细,有些部位破坏、断裂,骨髓腔明显扩大;补肾健骨中药复方组、仙灵骨葆阳性对照组与模型组相比,股骨头显微形态结构病理改变明显改善。③骨组织Leptin启动子-493bp~-19bp甲基化水平,模型组与正常组相比CpG19、CpG22.23显著升高(P<0.05);补肾健骨中药复方组(P<0.01)、仙灵骨葆阳性对照组(P<0.05)与模型组相比,CpG19甲基化水平显著降低。结论 补肾健骨中药复方通过降低骨组织Leptin启动子甲基化的分子机制,有效防治PMOP。 |

Objective To observe the methylation level of leptin promoter in bone of ovariectomized osteoporosis rats, and to explore the molecular mechanism of reinforcing kidney to strengthen bone Chinese herbal compound in preventing and treating postmenopausal osteoporosis (PMOP). Methods PMOP model was established by ovariectomy in Wistar rats. Rats were divided into 4 groups: normal group, model group, reinforcing kidney to strengthen bone Chinese herbal compound group, and Xianlinggubao positive control group. After 14 weeks of drug administration, bone mineral density was detected with X-ray absorptiometry. Micro-morphology structure of HE stained paraffin section of the femoral head was observed with the microscope. Leptin promoter methylation level was detected with MassARRAY. Results (1) Bone mineral density of the lumbar vertebrae 1-6 decreased significantly in model group compared to that in normal group (P<0.01) and increased significantly in reinforcing kidney to strengthen bone Chinese herbal compound group and Xianlinggubao positive control group compared to that in model group (P<0.05). (2) Compared to those in normal group, the cortical bone plate of the femoral head in model group became thinner, the cancellous bone trabeculae became obviously thinner with damage and fracture at some parts, and the bone marrow cavity was enlarged obviously in model group. The pathological changes of micro-morphology structure of the femoral head improved obviously in reinforcing kidney to strengthen bone Chinese herbal compound group and Xianlinggubao positive control group compared to those in model group. (3) The methylation level of leptin promoter -493bp - -19bp CpG19 and CpG22.23 in bone increased significantly in model group compared to that in normal group (P<0.05). The methylation level of leptin promoter -493bp - -19bp CpG19 in bone decreased significantly in reinforcing kidney to strengthen bone Chinese herbal compound group (P<0.01) and Xianlinggubao positive control group (P<0.05) compared with that in model group. Conclusion Reinforcing kidney to strengthen bone Chinese herbal compound reduces leptin promoter methylation in bone to prevent and treat PMOP effectively. |
