目的 研究补肾壮骨颗粒治疗绝经后骨质疏松症期间对子宫内膜及ERa、ERb、GPCR30的影响。方法 将60只6月龄SPF级SD雌性大鼠随机分为模型组、雷洛昔芬组、戊酸雌二醇组、中药组、假手术组,去卵巢建立绝经后骨质疏松症模型或行假手术。各组用药干预3个月后随机抽取半数测量骨密度,运用HE染色观察子宫组织形态结构并测量子宫内膜厚度、直径,通过Western blot 检测ERa、ERb、GPCR30的表达量。余下半数大鼠继续用药干预3个月(总用药6个月)并同样用上述方法处理检测。结果 中药组股骨及全身骨密度高于模型组,比较差异有统计学意义(P<0.05),子宫内膜厚度及直径大于模型组,但仅用药6个月的直径与MG的差异有统计学意义(P<0.05),ERa、ERb蛋白相对表达量高于模型组,差异有统计学意义(P<0.05),GPCR30蛋白相对表达量高于模型组,差异无统计学意义(P>0.05)。结论 补肾壮骨颗粒可以改善绝经后骨质疏松症导致的低骨量,同时子宫内膜无明显不良反应。 |

Objective To study the effect of Bushen Zhuanggu Granule on the expression of endometrium and ERa, ERb and GPCR30 in postmenopausal osteoporosis. Methods Sixty female Sprague-Dawley rats of 6 months old were randomly divided into sham operation group, model group, raloxifene group, estradiol valerate group and traditional Chinese medicine group. The rats in each group except sham operation group were established as postmenopausal osteoporosis model, and treated with drug intervention for 3 months, half of the rats in each group was randomly selected for checking the bone density, and the uterus tissue was preserved for HE to detect the endometrial thickness and diameter and for western blot that detect the expression of ERa, ERb and GPCR30. The remaining half of the rats continued to use the drug for 3 months, at the end, treated with the same method above. Results The femur and whole-body bone density of the Chinese medicine group were higher than that of the model group, the difference was statistically significant (P<0.05). The endometrial thickness and diameter were larger than the model group, but only the difference in diameter after 6 months of treatment was statistically significant (P<0.05). The relative expression of ERa and ERb protein was higher than that of the model group (P<0.05). There was no significant difference between the relative expression of GPCR30 protein and the model group (P>0.05). Conclusion Bushen Zhuanggu Granule can improve the low bone mass caused by postmenopausal osteoporosis and has no obvious adverse effects on the endometrium. |
