      目的 分析右归丸对骨性关节炎大鼠TGF-β1/Smads信号通路的影响。方法 将64只成年SD大鼠利用随机数字表法分为4组,每组15只。正常组不做任何处理;模型组、右归丸组、抑制剂组采用经典的Hulth法建立膝骨性关节炎模型,造模6周后,右归丸组灌服20g/(kg·d)的右归丸;抑制剂组腹腔注射100 μg/(kg·d)的TGF-β1/Smads信号通路抑制剂SB431542,同时灌服20 g/(kg·d)的右归丸;模型组灌服等剂量的生理盐水,连续给药8周。末次给药24 h后取关节软骨组织,制作病理切片观察软骨病理改变,采用ELISA法检测软骨组织中肿瘤坏死因子α与白细胞介素1β水平,免疫组化染色观察软骨组织中Ⅱ型胶原蛋白与基质金属蛋白酶13(MMP-13)阳性表达,采用RT-PCR法检测软骨组织中TGF-β1、Smad2、Smad3 mRNA表达,采用Western blot检测软骨组织中TGF-β1、p-Smad2、p-Smad3蛋白表达。结果 与模型组比较,抑制剂组软骨组织Mankin评分降低(P<0.05),软骨组织中肿瘤坏死因子α与白细胞介素1β水平降低(P<0.05),Ⅱ型胶原蛋白阳性表达升高(P<0.05),MMP-13阳性表达降低(P<0.05),TGF-β1、Smad2、Smad3 mRNA表达升高(P<0.05),TGF-β1、p-Smad2、p-Smad3蛋白表达升高(P<0.05)。与抑制剂组比较,右归丸组软骨组织Mankin评分降低(P<0.05),软骨组织中肿瘤坏死因子α与白细胞介素1β水平降低(P<0.05),Ⅱ型胶原蛋白阳性表达升高(P<0.05),MMP-13阳性表达降低(P<0.05),TGF-β1、Smad2、Smad3 mRNA表达升高(P<0.05),TGF-β1、p-Smad2、p-Smad3蛋白表达升高(P<0.05)。结论 右归丸对骨性关节炎大鼠软骨组织具有保护作用,其作用机制可能与TGF-β1/Smads信号通路有关。

      Objective To analyze the effect of Yougui Pill on TGF - β 1 / Smads signaling pathway in rats with osteoarthritis. Methods 64 adult SD rats were randomly divided into 4 groups with 15 rats in each group.The normal group did not do any treatment; the model group, Yougui Pill group and inhibitor group established knee osteoarthritis model by using the classic huith method. After 6 weeks of modeling, Yougui Pill group was given 20g / (kg · d) Yougui Pill; the inhibitor group was given 100 μ g / (kg · d) SB431542 intraperitoneally and 20g / (kg · d) Yougui Pill; the model group was given the same dose of normal saline for 8 consecutive weeks.24 hours after the last administration, articular cartilage tissue was taken and pathological sections were made to observe the pathological changes of cartilage. The concentrations of tumor necrosis factor α (TNF - α) and interleukin-1 β (IL-1 β) in cartilage tissue were detected by ELISA method. The positive expression of type II collagen and matrix metalloproteinase-13 (MMP-13) in cartilage tissue were observed by immunohistochemistry. TGF - β 1, Smad2 and Smad3 in cartilage tissue were detected by RT-PCR The expression of TGF - β 1, p-Smad2 and p-Smad3 in cartilage were detected by Western blot. Results Compared with the model group, the Mankin score of cartilage tissue in the inhibitor group was decreased (P < 0.05), the concentrations of tumor necrosis factor α and interleukin-1 β in cartilage tissue were decreased (P < 0.05), the positive expression of type II collagen was increased (P < 0.05), the positive expression of MMP-13 was decreased (P < 0.05), the mRNA expression of TGF - β 1, Smad2 and Smad3 was increased (P < 0.05), and the protein expressions of TGF - β 1, p-Smad2 and p-smad3 were increased (P < 0.05). Compared with the inhibitor group, the Mankin score of cartilage tissue in Yougui Pill group was decreased (P < 0.05), the concentration of tumor necrosis factor α and interleukin-1 β in cartilage tissue was decreased (P < 0.05), the positive expression of type II collagen was increased (P < 0.05), the positive expression of MMP-13 was decreased (P < 0.05), the mRNA expression of TGF - β 1, Smad2 and Smad3 was increased (P < 0.05), and the protein expressions of TGF - β 1, p-Smad2 and p-smad3 were increased (P < 0.05). Conclusion Yougui Pill has protective effect on cartilage tissue of osteoarthritis rats, and its mechanism may be related to TGF - β 1 / Smads signaling pathway.
