枸杞子治疗骨质疏松症的分子机制

      目的 运用网络药理学方法分析枸杞子治疗骨质疏松症的药理学机制。方法 从TCMSP分析平台上获取枸杞子药物的活性成分及潜在的作用靶点,运用Citespace3.7.1软件绘制“有效成分-靶点”网络图;同时分别在OMIM、Drugbank和Genecards三种数据库平台上获取骨质疏松疾病的相关基因,并取并集作为疾病的潜在作用靶点。将枸杞子和骨质疏松疾病的潜在作用靶点取交集,得到中药-疾病共同作用靶点。运用STRING数据分析平台对共同作用靶点进行PPI网络分析,利用cytohubba插件选取前20位显著性基因并取交集即可得到核心基因。对核心靶点进行GO功能和KEGG通路富分析,而后使用R语言软件对上述的富集结果进行可视化绘图。结果 共筛选获得枸杞子中主要有效成分36个,其中主要核心有效成分有槲皮素、黄豆黄素、β-谷固醇、豆甾醇、谷甾醇α1。活性成分所对应的靶点基因有117个,其中核心基因16个:IL-6、JUN、TNF等。GO富集所得的生物过程主要与调控成骨细胞和破骨细胞的增殖、分化有关。KEGG信号通路富集结果提示,枸杞子治疗骨质疏松疾病作用集中于IL-17、TNF、AGE-RAGE、MAPK等信号通路。结论 枸杞子可能通过多种有效成分、多靶点作用于免疫炎性反应、细胞分化等多方面来发挥抗骨质疏松症的作用。

      Objective To analyze the pharmacological mechanism of Lycii Fructus in the treatment of osteoporosis by using network pharmacology. Methods Obtained the active ingredients and potential targets of Lycii Fructus from the TCMSP analysis platform, and used Citespace 3.7.1 software to draw the "active ingredient-target" network diagram; meanwhile, the three database platforms of OMIM, Drugbank and Genecards Search for the keyword "osteoporosis" were obtained related genes of osteoporosis disease, and took the union as the potential target of the disease. The potential targets of Lycii Fructus and osteoporosis diseases were taken into intersection to obtain the common target of Chinese medicine-disease. Used the STRING platform to perform PPI network analysis on the common targets, and used the cytohubba to select the top 20 significant genes, and took the intersection to obtain the core genes. The GO function and KEGG pathway enrichment analysis of the core targets of Lycii Fructus and osteoporosis were performed through the Metasacpe database, and then the OmicShare cloud platform and R software were used to visualize the above enrichment results. Results A total of 36 main active ingredients in Lycii Fructuswere screened, and the main core active ingredients were quercetin, glycitein, β-sitosterol, stigmasterol, and sitosterol α1.There are 117 target genes corresponding to the active ingredients, including 16 core genes: IL-6, JUN, TNF, etc. The biological process obtained from GO enrichment was mainly related to the regulation of the proliferation and differentiation of osteoblasts and osteoclasts. The enrichment results of KEGG signaling pathway suggest that the effects of Lycii Fructusfruit in the treatment of osteoporosis were focused on the signaling pathways such as IL-17, TNF, AGE-RAGE, and MAPK. Conclusion Lycii Fructus may exert its anti-osteoporosis effect through multiple active ingredients and multiple targets acting on immuno-inflammatory response and cell differentiation.
