Objective To study and explores the effect of Xianling Gubao Capsule on the expression of bone growth factor BMP-2 and IGF-1 and fracture healing in rats with osteoporotic fracture. Methods 48 female SD rats were randomly divided into: Sham operation group, Model group, Estradiol group, Xianling Gubao group, 12 rats/group, "Bilateral oophorectomy + intramedullary fixation of right femoral shaft fracture" was used to construct a rat model of osteoporotic fracture, Assess fracture healing, detect femoral callus BMD, femoral bone biomechanics indicators and serum bone metabolism related indicators, and detect the protein expression of callus BMP-2 and IGF-1. Detection of femoral callus BMD, femoral bone biomechanics indexes and serum bone metabolism related indexes in each group, Immunohistochemical detection of BMP-2 and IGF-1 protein expression in callus of each group. Results Compared with the sham operation group, fracture healing score, femoral callus BMD, femoral bone biomechanics indexes (maximum load, maximum stress, maximum displacement), callus BMP-2 and IGF-I positive expression were significantly lower than the model group (P<0.05), Compared with the model group, the estradiol group and the Xianling Gubao group had significantly higher fracture healing scores, femoral callus BMD, femoral bone biomechanics indexes, callus BMP-2 and IGF-I positive expressions (P<0.05), Xianling Gubao group is the highest. The serum bone metabolism indexes (BGP, PICP, TRACP-5b) of the model group were significantly higher than the sham operation group (P<0.05), The serum bone metabolism indexes of the estradiol group and the Xianling Gubao group were significantly lower than the model group (P<0.05), and the Xianling Gubao group was the lowest. Conclusion Xianling Gubao Capsule may mediate to increase the expression of bone growth factor BMP-2 and IGF-1 in rats with osteoporotic fractures, improve bone metabolism, accelerate callus formation, increase bone density, improve bone biomechanics, and promote fracture healing. |
