      目的 观察淫羊藿苷(icariin,ICA)与雌激素(estrogen,E2)对SD大鼠骨髓间充质干细胞(bone marrow mesenchymal stem cells,BMSCs)向成骨分化及对内环境雌激素水平的影响。方法 取3、4代BMSCs细胞,分为正常组、E2组、ICA组和E2+ICA组,在37 ℃ CO2培养箱内诱导18 d;运用MTT法检测培养24、48、72 h后各组细胞增殖;通过ELISA检测法检测E2及6、12、18 d时的碱性磷酸酶(alkaline phosphatase,ALP)活性;运用茜素红染色法观察各组成骨分化情况;运用免疫荧光法观察雌激素受体α(estrogen receptor-α,ERα)的表达与分布。结果 根据MMT结果,正常组、E2组、ICA组和E2+ICA组均可促进BMSCs增殖(P<0.01);与正常组相比,各给药组ALP活性升高(P<0.01)。相同时间点各给药组ALP活性从小到大依次为ICA组<E2组<E2+ICA组,12 d时各组浓度达到峰值;同期观测12 d时茜素红染色及免疫荧光染色结果(P<0.05),与ALP结果趋势一致;各给药组E2活性明显升高(P<0.01),从小到大依次为ICA组<E2+ICA组<E2组,与ALP结果趋势不一致。结论 ①E2+ICA组明显优于单独给药组。与E2相比ICA反应更快、毒性更小;②ICA可双向调节内环境雌激素的含量,使其趋于稳态,以规避雌激素替代疗法(estrogen replacement therapy ,ERT)副作用;③ICA能够通过调节雌激素水平,提高ERα核内表达,增加ALP含量,可用于治疗以雌激素缺乏为主的绝经后骨质疏松症,亦可为绝经后雌激素分泌异常导致的其他慢性病提供新思路。

      Objective To observe the effect of icariin (ICA) and estrogen (E2) on the osteogenic differentiation of bone marrow mesenchymal stem cells (BMSCs) of SD rats and the level of estrogen in the internal environment Impact. Methods The 3rd and 4th passages of BMSCs were divided into normal group, E2 group, ICA group and E2+ICA group. Induced 18 days in a CO2 incubator at 37℃; used MTT method to detect cell proliferation after 24, 48, and 72 hours of culture;used ELISA method to detect E2 and alkaline phosphatase (ALP) at 6, 12, and 18 days Activity; used Alizarin Red staining method to observe the differentiation of bones; used immunofluorescence method to observe the expression and distribution of estrogen receptor-α (ERα). Results The E2 group, ICA group, and E2 + ICA group promoted BMSCs proliferation by MTT(P<0.01) and increased ALP activity in each experimental group compared to normal group ( P<0.01). At the same time point, the trend of ALP activity from small to largewas ICA group<E2 group<E2 + ICA group,The concentration of each group reached the peak at 12d.The results of Alizarin Red staining and immunofluorescence staining on the 12th day of observation at the same time (P<0.05) were consistent with the trend of ALP results. E2 activity of each experimental group was increased (P<0.01), the trend of E2 activity from small to large was ICA group<estrogen+ ICA group<estrogen group but it is different from the trend of ALP results. Conclusion ①The E2 + ICA group is significantly superior to the drug group alone.ICA responded faster and is less toxic compared to E2.② ICA can bidirectional regulate the content of environmental estrogen to its homeostasis to circumvent ERT side effects.③ICA can regulate the level of estrogen, increase the expression of ERα in the nucleus, and increase the content of ALP. It can be used to treat postmenopausal osteoporosis that is dominated by estrogen deficiency. It can also provide new ideas for other chronic diseases caused by abnormal estrogen secretion after menopause.
