      目的 探讨不同代数骨髓源性巨噬细胞增殖及诱导破骨分化的规律。方法 选取8周龄C57BL/6小鼠P0~P4共5代骨髓源性巨噬细胞,随机选择镜下视野计数杂质细胞与衰老细胞;运用CCK8法检测骨髓源性巨噬细胞的增殖能力;诱导巨噬细胞破骨分化,通过抗酒石酸酸性磷酸酶染色量化破骨细胞数量与破骨细胞面积,比较不同代数之间指标差异。 结果 P0组、P1组骨髓源性巨噬细胞的杂质较P2组、P3组、P4组更多,且差异有统计学意义;96 h内,P2组骨髓源性巨噬细胞的增殖能力最强,P3组、P4组次之,P0组、P1组最差;诱导破骨分化后,P2组骨髓源性巨噬细胞诱导出破骨细胞数量最多,面积最大,P3组次之,P0组、P1组、P4组诱导出破骨细胞数量少,面积小。结论 P2组骨髓源性巨噬细胞的纯度最高,增殖速度最快,破骨分化能力最强。

      Objective To investigate the rules of proliferation and osteoclast differentiation induced by different algebraic macrophages. Methods Five generations of bone-marrow-derived macrophages were selected from P0-P4 of 8-week-old C57BL/6 mice, and impurity cells and senescent cells were counted by random selection under the microscope field of vision. CCK8 assay was used to detect the proliferation ability of bone marrow derived macrophages. To induce osteoclast differentiation of macrophages, the number of osteoclasts and the area of osteoclasts were quantified by tartrate-resistant acid phosphatase staining, and the index differences between different algebrae were compared. Results The impurities of bone marrow-derived macrophages in P0 group and P1 group were more than those in P2 group, P3 group and P4 group, and the difference was statistically significant. Within 96h, the proliferation ability of bone marrow-derived macrophages in group P2 was the highest, followed by that in group P3 and P4, and that in group P0 and P1 was the worst. After the induction of osteoclast differentiation, the number and area of osteoclasts induced by bone-marrow-derived macrophages in group P2 were the largest, followed by those in group P3, and the number and area of osteoclasts induced in group P0, P1 and P4 were small. Conclusion The purity of bone marrow-derived macrophages is the highest, the proliferation rate is the fastest, and the ability of osteoclastic differentiation is the strongest in P2 group.
